Bortezomib (Velcade®) is a first‑in‑class proteasome inhibitor that was initially approved by the U.S. Food and Drug Administration in 2003 for the treatment of multiple myeloma, initiating a major shift toward targeted therapy in hematologic cancers. By blocking the 26S proteasome, it disrupts protein degradation, leading to cancer cell apoptosis, which was an entirely novel mechanism at the time of its approval.

Real-world data show that GLP-1 receptor agonists (GLP-1s) may reduce metastatic progression of certain obesity-related cancers, namely lung, breast, colorectal, and liver cancers. In addition, GLP-1 receptor (GLP-1R) expression is associated with overall survival, suggesting that GLP-1 signaling could be involved in the progression of these cancers.
