Survival outcomes with matched targeted therapy in advanced biliary tract cancer: a large global cohort analysis
Abstract The prognosis of advanced biliary tract cancer remains poor despite up to 40% of tumors harboring actionable molecular alterations. This international collaborative study from Mayo Clinic, Duke Cancer Institute, National Cancer Center East (Japan), and the SCRUM-Japan GOZILA/MONSTAR-SCREEN projects evaluated clinical outcomes in 1049 patients with advanced BTC based on tumor molecular profiling and matched treatment selection.
Pharmacologic features, clinical applications, and drug safety evaluation of futibatinib in the treatment of biliary tract cancer (BTC)
ABSTRACT Introduction Futibatinib is a small, potent, covalent, irreversible fibroblast growth factor receptor (FGFR) 1–4 inhibitor that has been added as a new standard of care for previously treated unresectable and/or advanced FGFR2 fusion/rearrangement-positive BTC. FGFR2 fusions/rearrangements play a key role in BTC survival, proliferation, invasion, and development of distant metastasis. The inhibition of this pathway is an important target in the treatment of BTC.
Advances in Targeting KRAS G12C in Gastrointestinal Malignancies: Focus on KRAS G12C Inhibitors
Overview Kirsten rat sarcoma virus (KRAS) gene mutations are amongst the most prevalent oncogenic mutations in gastrointestinal tumours. Attempts at targeting the KRAS protein have been unsuccessful for decades, mainly due to its challenging architecture that lacks small-molecule-binding sites.
