19F-PCS measurements on proteins in live mammalian cells
19F-PCS measurements on proteins in live mammalian cells Pseudocontact shift (PCS) contains rich structural information of proteins in structural and chemical biology. Here, 19F-PCS is achieved in live mammalian cells via dual labelling of the target protein with a paramagnetic tag and a 19F-tag. About 0.1 ppm 19F-PCS can be determined over 20 Å from the paramagnetic center in live cells. This is the first obervation 19F-PCS in live mammalian cells.
Pt(IV) complex selectively oxidizes alpha-synuclein methionine as disclosed by NMR
Pt(iv) complex selectively oxidizes alpha-synuclein methionine as disclosed by NMR† Bin-Bin Pan,a Hui-Zhong Liu,a Wei-Han Menga and Xun-Cheng Su *a Abstract Alpha-synuclein (αS) is an intrinsically disordered neuronal protein, and the oxidative modification of αS promotes its oligomerization and accumulation in Parkinson's disease.
Pt(IV) Complex Selectively Oxidizes alpha-Synuclein Methionine Disclosed by NMR
Pt(IV) Complex Selectively Oxidizes alpha-Synuclein Methionine Disclosed by NMR Xun-Cheng Su, Bin-Bin Pan, Hui-Zhong Liu and Wei-Han Meng Abstract Alpha-synuclein (αS) is an unstructured neuronal protein, and its oxidative modification promotes oligomerization and accumulation in Parkinson's disease. In this study, we characterize methionine oxidation in αS at residue level by incubating with a redox-active platinum(IV) by nuclear magnetic resonance (NMR) and mass spectrometry.
Third BIR domain of XIAP binds to both Cu(II) and Cu(I) in multiple sites and with diverse affinities characterized at atomic resolution
Abstract The X-chromosome linked inhibitor of apoptosis, XIAP, is mainly known as the inhibitor of caspases by direct interaction with caspases with its baculoviral IAP repeat (BIR) domains. XIAP has three BIR domains and each BIR domain contains a zinc binding site, normally known as zinc finger motif. Recent studies showed that XIAP is involved in copper homeostasis in cells and the BIR domains bind copper ion.
