Lymph node-targeted, mKRAS-specific amphiphile vaccine in pancreatic cancer: the randomized phase 2 AMPLIFY-7P trial
Abstract Camupepimut (ELI-002 7P) drives mutated KRAS (mKRAS)-specific T cell responses that correlated with disease-free survival (DFS) and overall survival (OS) in patients with mKRAS pancreatic adenocarcinoma (PDAC) in the phase 1 AMPLIFY-201 trial. In the phase 2 AMPLIFY-7P trial, patients with stage I-III KRAS-mutated PDAC without recurrence within 6 months of standard therapy were randomized 2:1 to camupepimut (n = 96) or standard-of-care/observation (n = 48).

Addition of High-Dose Vitamin D3 to Standard Treatment in Patients With Metastatic Colorectal Cancer: The SOLARIS Randomized Clinical Trial (Alliance A021703) Key Points Question Does the addition of high-dose vitamin D3 to standard treatment improve progression-free survival in patients with metastatic colorectal cancer?

Lymph node-targeted, mKRAS-specific amphiphile vaccine in pancreatic and colorectal cancer: phase 1 AMPLIFY-201 trial final results - Nature Medicine
Abstract Cellular immunity, mediated by tumor antigen-specific CD4+ and CD8+ T cells, has a critical role in the success of cancer immunotherapy by targeting intracellular driver and passenger tumor mutations.
Lymph-node-targeted, mKRAS-specific amphiphile vaccine in pancreatic and colorectal cancer: the phase 1 AMPLIFY-201 trial - Nature Medicine
Abstract Pancreatic and colorectal cancers are often KRAS mutated and are incurable when tumor DNA or protein persists or recurs after curative intent therapy. Cancer vaccine ELI-002 2P enhances lymph node delivery and immune response using amphiphile (Amph) modification of G12D and G12R mutant KRAS (mKRAS) peptides (Amph-Peptides-2P) together with CpG oligonucleotide adjuvant (Amph-CpG-7909).

Phase 1 study of the ATR inhibitor berzosertib in combination with cisplatin in patients with advanced solid tumours
Abstract Background Berzosertib (formerly M6620, VX-970) is a highly potent and selective, first-in-class ataxia telangiectasia-mutated and Rad3-related protein kinase (ATR) inhibitor. We assessed the safety, tolerability, pharmacokinetics, and preliminary efficacy of berzosertib plus cisplatin. Methods Adult patients with advanced solid tumours refractory or resistant to standard of care therapies received ascending doses of cisplatin (day 1) and berzosertib (days 2 and 9) every 3 weeks (Q3W).
Simple parameters to solve a complex issue: predicting response to checkpoint inhibitor therapy in lung cancer
Immune checkpoint inhibitors (ICIs) have transformed the standard of care of non-small-cell lung cancer (NSCLC) over recent years. Based on large prospective clinical trials, pembrolizumab, a programmed death-1 (PD-1) inhibitor, received US FDA approval as first-line therapy as both a single agent in patients with PD-L1-expressing tumors and in combination with chemotherapy for all NSCLC patients regardless of programmed death ligand 1 (PD-L1) status [1–4].
Pretreatment nutritional status and response to checkpoint inhibitors in lung cancer
Practice points Out of the 106 patients analyzed, 17% of patients had a low albumin (<3.5 g/dl) at baseline and 37.7% reported weight loss of 5% or more in the 6 months prior to starting immunotherapy.
