Mapping heterogeneity drivers in human PSC-derived limbal stem cell differentiation through single-cell multi-modal analysis
Abstract Understanding the molecular underpinnings of stem cell differentiation is pivotal for generating appropriate cell types in cell-based therapies. Differentiation of human pluripotent stem cells (PSC) into corneal limbal stem cells offers a promising avenue to regenerate the corneal epithelium.
Metabolic and neuroactivity imbalances in plasma from aniridia patients with PAX6 haploinsufficiency
Abstract PAX6 is a transcription factor crucial for the development of the eye, pancreas, and brain. Heterozygous variants resulting in PAX6 haploinsufficiency are the main genetic cause of congenital aniridia, characterized by both anterior and posterior ocular defects and sight loss. The extra-ocular features of PAX6 haploinsufficiency are becoming more widely recognised, with systemic manifestations like obesity, diabetes, and neurological/behavioural disorders being reported.
Deciphering the heterogeneity of differentiating hPSC-derived corneal limbal stem cells through single-cell RNA sequencing
Highlights • hPSCs to LSCs span epithelial, mesodermal, and undifferentiated cell states • scRNA-seq reveals the cell line-dependent differentiation heterogeneity • ITGA6 and AREG can be used to select pure LSC-like subpopulation Summary A comprehensive understanding of the human pluripotent stem cell (hPSC) differentiation process stands as a prerequisite for the development of hPSC-based therapeutics.
Deciphering the heterogeneity of differentiating hPSC-derived corneal limbal stem cells through single-cell RNA-sequencing
Abstract A comprehensive understanding of the human pluripotent stem cell (hPSC) differentiation process stands as a prerequisite for the development hPSC-based therapeutics. In this study, single-cell RNA-sequencing (scRNA-seq) was performed to decipher the heterogeneity during differentiation of three hPSC lines towards corneal limbal stem cells (LSCs).
Identification of the regulatory circuit governing corneal epithelial fate determination and disease
Materials and methods All procedures for establishing and maintaining human primary keratinocytes were approved by the ethical committee of the Radboud University Medical Center (“Commissie Mensgebonden Onderzoek Arnhem-Nijmegen”) (CMO-nr:2004/132). All donors have given informed consent in written forms. All procedures for establishing and maintaining LSCs were conducted according to the principles expressed in the Declaration of Helsinki.
Single-Cell RNA Sequencing: Opportunities and Challenges for Studies on Corneal Biology in Health and Disease
Abstract : The structure and major cell types of the multi-layer human cornea have been extensively studied. However, various cell states in specific cell types and key genes that define the cell states are not fully understood, hindering our comprehension of corneal homeostasis, related diseases, and therapeutic discovery. Single-cell RNA sequencing is a revolutionary and powerful tool for identifying cell states within tissues such as the cornea.
