Here, we primarily explored the influence of Cre-driver strain on Cre-mediated recombination and mosaicism at a fixed locus—R26. We evaluated three widely used Cre-driver strains with robust promoters for global gene knockout: 1) Ella-cre, 2) CMV-cre, and 3) Sox2-cre (Fig. 2a). The Ella-cre driver strain served as a positive control, based on published data indicating that this strain mediates widespread mosaicism [41].
