In vivo biomarkers of GABAergic function in epileptic rats treated with the GAT‐1 inhibitor E2730
E2730 demonstrated powerful anti-seizure effects in the kainic acid status epilepticus (KASE) rat model of chronic drug-resistant mesial temporal lobe epilepsy (MTLE), with a high proportion of animals being rendered seizure-free. E2730 treatment reduced brain taurine levels measured by magnetic resonance spectroscopy (MRS) in KASE and control rats, whereas [18F]Flumazenil positron emission tomography (PET) imaging revealed no changes in γ-aminobutyric acid (GABA) receptor binding.

Epilepsy phenotype and its reproducibility after lateral fluid percussion‐induced traumatic brain injury in rats: Multicenter EpiBioS4Rx study project 1
The first large-scale preclinical multicenter study in epilepsy Epilepsy phenotype after lateral fluid percussion-induced traumatic brain injury is highly reproducible across different study sites Preclinical multicenter studies are feasible 1 INTRODUCTION Traumatic brain injury (TBI) is defined as an alteration in brain function, or other evidence of brain pathology, caused by an external force.1 Annually, approximately 2.5 million people in both Europe and the USA experience TBI...
E2730, an uncompetitive γ‐aminobutyric acid transporter‐1 inhibitor, suppresses epileptic seizures in a rat model of chronic mesial temporal lobe epilepsy
• There are dose-dependent plasma levels of E2730 in healthy rats during continuous infusion via osmotic pumps • Continuous infusion with E2730 results in no/minimal sedative or neuromotor adverse effects at antiseizure doses • Continuous infusion with E2730 results in strong seizure suppression in a model of chronic TLE 1 INTRODUCTION Epilepsy is a neurological disorder that is characterized by recurrent unprovoked seizures.
