Structural heterogeneity of uncrosslinked FtsH-HflKC assemblies includes variable FtsH occupancy and a distinct wild-type FtsH architecture
Abstract The AAA+ metalloprotease FtsH associates with HflK and HflC to form the megadalton FtsH-HflKC complex (FHC), which contributes to membrane-protein quality control in Escherichia coli. Recent structural studies have revealed asymmetric HflKC architectures and variable FtsH occupancy, contrasting with earlier crosslinked structures of a symmetric four-hexamer cage.
Allosteric modulation by the fatty acid site in the glycosylated SARS-CoV-2 spike
Abstract The trimeric spike protein plays an essential role in the SARS-CoV-2 virus lifecycle, facilitating virus entry through binding to the cellular receptor angiotensin-converting enzyme 2 (ACE2) and mediating viral and host membrane fusion. The SARS-CoV-2 spike contains an allosteric fatty acid (FA) binding site at the interface between two neighbouring receptor-binding domains.
Evaluation and deployment of isotype-specific salivary antibody assays for detecting previous SARS-CoV-2 infection in children and adults - Communications Medicine
Abstract Background Saliva is easily obtainable non-invasively and potentially suitable for detecting both current and previous SARS-CoV-2 infection, but there is limited evidence on the utility of salivary antibody testing for community surveillance. Methods We established 6 ELISAs detecting IgA and IgG antibodies to whole SARS-CoV-2 spike protein, to its receptor binding domain region and to nucleocapsid protein in saliva.
Bottom-up assembly of viral replication cycles - Nature Communications
Towards the bottom-up assembly of a compartmentalized virus-like structure capable of intracellular replication, certain essential features of the virions of choice need to be considered for the design process: Virions have evolved as highly efficient carriers of genetic information that have mastered the reprogramming of host cells for viral propagation. As inherent infectious agents, viruses exploit the molecular machinery of host cells to produce and propagate viral particles.
PhD Position in Next-Generation Vaccine Engineering (Sept. 2018) !
The position is based on ADDomer, a novel protein scaffold we developed, which is uniquely suited for developing next-generation vaccines. The project combines cutting-edge protein engineering, state-of-the-art Cryo-EM, and using our MultiBac technology platform.
