Long-Read RNA-sequencing reveals transcript-specific regulation in human-derived cortical neurons
Abstract Long-read RNA sequencing has transformed transcriptome analysis by enabling comprehensive mapping of full-length transcripts, providing an unprecedented resolution of transcript diversity, alternative splicing, and transcript-specific regulation.
Unraveling Axonal Transcriptional Landscapes: Insights from iPSC-Derived Cortical Neurons and Implications for Motor Neuron Degeneration
Abstract Neuronal function and pathology are deeply influenced by the distinct molecular profiles of the axon and soma. Traditional studies have often overlooked these differences due to the technical challenges of compartment specific analysis. In this study, we employ a robust RNA-sequencing (RNA-seq) approach, using microfluidic devices, to generate high-quality axonal transcriptomes from iPSC-derived cortical neurons (CNs).
A transcriptomic atlas of aged human microglia
The biological specimens used in this study originated from two prospective studies of aging: the Religious Orders Study (ROS)6 and the Memory and Aging Project (MAP)7. Participants have to be at least 53 (ROS) or 55 (MAP) years old and non-demented at enrollment and sign an Anatomical Gift Act agreeing to donate their brain and spinal cord at the time of death. Thus, they represent a sample of the older population from the Chicago, IL metropolitan area.
A human microglia-like cellular model for assessing the effects of neurodegenerative disease gene variants
In a new study, Ryan et al. characterized a cellular model of monocytes differentiated into a microglial-like state. They then demonstrated the effectiveness of this human in vitro cell model system for identifying functional consequences of genetic variation associated with neurodegenerative disease risk by performing a cis-expression quantitative trait locus (cis-eQTL) study.
Genetic epistasis regulates amyloid deposition in resilient aging
Cognitive decline associated with pathological burden in primary age-related tauopathy
