LUBAC PUB domain interactions restrict Met1-linked ubiquitination to prevent murine embryonic lethality and immune pathology
Abstract Met1-linked ubiquitin assembled by LUBAC coordinates immune receptor signalling and inflammation. The PUB domain of the LUBAC subunit HOIP interacts with PUB-interacting motif-containing proteins, including the deubiquitinases OTULIN and SPATA2-CYLD. Yet, how these interactions regulate LUBAC remains unclear. Here, we investigate the consequences of ablating PUB-interacting motif interactions by generating mice carrying an N101D mutation in the HOIP PUB domain.
The Met1-Ub System in T Cells: What We Know, What We Think We Know
1 Introduction Ubiquitination refers to the post-translational modification of proteins (and non-protein substrates) by one or more ubiquitin molecules, a process that regulates most cellular functions and signalling pathways [1]. Ubiquitin (Ub) is a small protein that encompasses 76 amino acids, of which the N-terminal methionine (Met1) and the seven lysine (Lys) residues serve as the attachment point for subsequent Ub moieties to generate differently linked polymeric Ub (polyUb) chains.
The Wrong Door Into the Medicine Cabinet
Somewhere in America this morning, someone injected a substance into an aching shoulder that has never been studied in a human being. The substance is TB-5001,2 one of a family of synthetic peptides that have become a phenomenon in gyms, wellness clinics and longevity podcasts. Another, BPC-157, appears in more than 50 million YouTube and TikTok videos3 promoted as a cure for torn tendons and inflamed guts. Vials arrive stamped "for research use only," a label that fools no one4.

M1-linked ubiquitination by LUBAC regulates AMPK signalling and the response to energetic stress - Cell Death & Differentiation

Abstract Methionine-1 (M1)–linked ubiquitin chains, assembled by the linear ubiquitin chain assembly complex (LUBAC) and disassembled by the deubiquitinase OTULIN, are critical regulators of inflammation and immune homoeostasis. Genetic loss or mutation of the LUBAC subunits HOIP and HOIL-1 or of OTULIN causes autoinflammatory syndromes accompanied by metabolic defects, including amylopectinosis, lipodystrophy, and fatty liver disease.

Multi-level orchestration of γδ T cell development and central nervous system inflammation by retinoic acid
Abstract The vitamin A metabolite retinoic acid (RA) is critical for the maturation and function of the immune system, however, our knowledge regarding its role in gamma delta (γδ) T cells is limited.

Clinical Characteristics and Pharmacological Management of COVID-19 Vaccine-Induced Immune Thrombotic Thrombocytopenia With Cerebral Venous Sinus Thrombosis: A Review | Coagulation Disorders | JAMA Cardiology | JAMA Network
Figure 1. Schematic Representation of the Pathogenesis of Vaccine-Induced Immune Thrombotic Thrombocytopenia (VITT) and Therapeutic Targets Owing to lack of high-quality data, management recommendations are based on expert opinion and extrapolation from treatment of heparin-induced thrombocytopenia given similarities between these 2 conditions.
Low-dose aspirin for early COVID-19: does the early bird catch the worm?
1. Low-dose aspirin for early COVID-19 During the ongoing COVID-19 pandemic caused by SARS-CoV-2 virus, the vast majority of patients have a mild to moderate illness that does not necessitate hospitalization and are appropriately treated in an ambulatory setting. Optimization of the outpatient treatment can potentially reduce viral spread and in hospitals reaching full capacity.

Pharmaco-Immunomodulatory Therapy in COVID-19
Abstract The severe acute respiratory syndrome coronavirus 2 associated coronavirus disease 2019 (COVID-19) illness is a syndrome of viral replication in concert with a host inflammatory response. The cytokine storm and viral evasion of cellular immune responses may play an equally important role in the pathogenesis, clinical manifestation, and outcomes of COVID-19.
The immune checkpoint receptor associated phosphatases SHP‐1 and SHP‐2 are not required for γδT17 cell development, activation or skin inflammation
