Hypoferremic Response to Chronic Inflammation Is Controlled via the Hemojuvelin/Hepcidin/Ferroportin Axis and Does Not Involve Hepcidin‐Independent Regulation of Fpn mRNA
1 Introduction Systemic iron traffic is subjected to hormonal regulation by hepcidin, a peptide secreted from the liver that targets the iron exporter ferroportin in intestinal epithelial cells, tissue macrophages, hepatocytes, and other cell types [1]. The binding of hepcidin occludes ferroportin's iron-efflux channel and promotes internalization and lysosomal degradation of the protein, thereby inhibiting iron entry into the bloodstream and eventually resulting in tissue iron retention.
Nutritional Aspects of Iron in Health and Disease
Abstract : Dietary iron assimilation is critical for health and essential to prevent iron-deficient states and related comorbidities, such as anemia. The bioavailability of iron is generally low, while its absorption and metabolism are tightly controlled to satisfy metabolic needs and prevent toxicity of excessive iron accumulation. Iron entry into the bloodstream is limited by hepcidin, the iron regulatory hormone.
Iron overload inhibits BMP/SMAD and IL-6/STAT3 signaling to hepcidin in cultured hepatocytes

It is generated in hepatocytes mainly in response to increased body iron stores or inflammatory cues.
