Coupled Solvent and Protein Dynamics Confer Differences in Exon-19 Deletion Mutants of the Epidermal Growth Factor Receptor Kinase
Abstract Deletions in Exon-19 of the epidermal growth factor receptor (EGFR) play a pivotal role in the pathogenesis of non-small cell lung cancer (NSCLC), influencing patient response to tyrosine kinase inhibitors (TKIs). Although these mutations are known to affect treatment efficacy, the precise molecular mechanisms have been unclear.
EGFR and tyrosine kinase inhibitor interactions probed by hydrogen-deuterium exchange and mass spectrometry (HDX-MS)
Abstract EGFR is one of the primary drug targets for treating non-small cell lung cancer (NSCLC) patients carrying EGFR oncogenic mutations in the tyrosine kinase domain (TKD). Such patients typically receive tyrosine kinase inhibitors (TKIs) to inhibit aberrant activation of EGFR; however, together with the appearance of the TKI-resistant mutations, TKIs' severe side effects often limit their clinical usage.
Coupled Solvent Dynamics and Protein Dynamics Help Drive Functional Differences in Exon-19 Deletion Mutants in the Epidermal Growth Factor Receptor (EGFR/ErbB1/HER1) Kinase Domain
Biochemical and structural basis for differential inhibitor sensitivity of EGFR with distinct exon 19 mutations - Nature Communications
Abstract Tyrosine kinase inhibitors (TKIs) are used to treat non-small cell lung cancers (NSCLC) driven by epidermal growth factor receptor (EGFR) mutations in the tyrosine kinase domain (TKD). TKI responses vary across tumors driven by the heterogeneous group of exon 19 deletions and mutations, but the molecular basis for these differences is not understood. Using purified TKDs, we compared kinetic properties of several exon 19 variants.
Enantioselective Halofunctionalization of Alkenes
Enantioselective Halofunctionalization of Alkenes PDF ShareShare Abstract Asymmetric functionalization of alkenes is a powerful transformation to access a variety of chiral molecules. This chapter discusses strategies for the reagent‐controlled enantioselective chloro‐, bromo‐, or iodofunctionalization of alkene substrates.
