Obtaining Valid Compatibility Intervals for Sequence Symmetry Analyses Utilizing Active Comparators: A Simulation Study
Using an active comparator in sequence symmetry analysis reduces bias from confounding by indication. The optimal estimator for calculating 95% compatibility intervals for active comparator sequence ratios is not known. The Miettinen-Nurminen score estimator achieved the most accurate 95% compatibility interval coverage of the tested estimators. The Miettinen-Nurminen score estimator provides the most accurate coverage, particularly in scenarios with low sample sizes.

Fathers of twins have indication of a slightly increased risk of postpartum depression around 6 months postpartum compared to fathers of singletons.
The Spironolactone Initiation Registry Randomized Interventional Trial in Heart Failure with Preserved Ejection Fraction (SPIRRIT‐HFpEF): Rationale and design
Heart failure with preserved ejection fraction and potential for mineralocorticoid receptor antagonists Heart failure (HF) with preserved ejection fraction (HFpEF) is a major and growing public health concern with limited treatment options.1-4 HFpEF is defined as HF with a left ventricular ejection fraction (LVEF) ≥50%.1 HF with a reduced ejection fraction has previously been defined as LVEF <40% and more recently ≤40%.
Response to letter about “Potential mechanisms of acyl‐ghrelin in heart failure: insights into cholinergic modulation”
We thank Dr Hrabovska for providing important additional mechanistic insights into the beneficial effects of acyl ghrelin in heart failure (HF).1 In our original randomized trial in patients with HF with reduced ejection fraction (HFrEF), by Lund et al., intravenous acyl ghrelin for 2 h increased cardiac output (CO) by 28% without lowering blood pressure, increasing heart rate, or causing arrhythmias or ischemia.2 In a secondary analysis, we showed that right ventricular-pulmonary arterial...
Balcinrenone plus dapagliflozin in patients with heart failure and chronic kidney disease: Results from the phase 2b MIRACLE trial
Introduction In patients with heart failure (HF), concomitant renal dysfunction is common and negatively affects prognosis and complicates HF treatment.1 Although mineralocorticoid receptor (MR) antagonists are a foundational therapy for HF with reduced ejection fraction, they are substantially underused in patients with chronic kidney disease (CKD)2 and are not recommended in those with an estimated glomerular filtration rate (eGFR) <30 ml/min/1.73 m2.3, 4 Balcinrenone (previously AZD9977)...

Epidemiology, Clinical Characteristics and Cause-specific Outcomes in Heart Failure with Preserved Ejection Fraction
Average (ratings) Abstract Heart failure (HF) is a global pandemic affecting 64 million people worldwide. HF with preserved ejection fraction (HFpEF) has traditionally received less attention than its main counterpart, HF with reduced ejection fraction (HFrEF). The incidence and prevalence of HFpEF show geographic variation and are increasing over time, soon expected to surpass those of HFrEF. Morbidity and mortality rates of HFpEF are considerable, albeit lower than those of HFrEF.

1 INTRODUCTION Individuals with haematological malignancies are at an increased risk of adverse outcomes following SARS-CoV-2 infection, with reported case fatality risks (CFR) ranging between 14% and 70%.1 A Danish study based on 108 individuals reported a 90-day mortality of up to 28%,2 while an English study based on 128 patients reported sequentially falling 90-day mortality from 42% to 2% over the course of the pandemic.3 Existing studies on the CFR of SARS-CoV-2 infection among...
A case for screening real‐world data for collateral drug benefits: Glucagon‐like peptide 1 receptor agonists and bile acid diarrhea
Abstract Purpose Collateral drug benefits are hitherto unknown beneficial effects that might lead to repurposing of already marketed drugs. A randomized controlled trial has found liraglutide to be non-inferior to colesevelam in reducing bile acid diarrhea. We hypothesized that this collateral drug benefit of liraglutide could have been detected using observational data. Methods We performed a sequence symmetry analysis (SSA).
Global Public Health Burden of Heart Failure: An Updated Review
Heart failure (HF) is a clinical syndrome with different underlying aetiologies rather than being a specific disease. Traditionally, HF has been defined as a condition where there is a reduced ability of the heart to pump and/or fill with blood, or alternatively an inadequate cardiac output caused by a structural or functional abnormality, or adequate cardiac output secondary to compensatory neurohormonal activation and increased left ventricular filling pressure.
Myeloperoxidase Inhibition Reverses Biomarker Profiles Associated With Clinical Outcomes in HFpEF:
JACC Journals › JACC: Heart Failure › Archives › Vol. 11 No. 7 Abstract Background Systemic microvascular dysfunction and inflammation are postulated to play a pathophysiologic role in heart failure with preserved ejection fraction (HFpEF). Objectives This study aimed to identify biomarker profiles associated with clinical outcomes in HFpEF and investigate how inhibition of the neutrophil-derived reactive oxygen species–producing enzyme, myeloperoxidase, affects these biomarkers.

SuPARnostic: An Advanced Predictive Tool for Detecting Recurrence in Renal Cell Carcinoma
BACKGROUND AND AIMS Renal cell carcinoma (RCC) is the most common malignant kidney tumour, with clear cell RCC (ccRCC) accounting for 70–80% of all cases.1 Approximately 15–20% of patients have primary metastatic RCC at diagnosis, and 15–20% of those who receive curative treatment for localised tumours will experience recurrence within 5 years of follow-up.2 Despite standard radiological imaging follow-up protocols, 30% of recurrences are found outside these protocols, and only 10% of...
Cox regression using a calendar time scale was unbiased in simulations of COVID-19 vaccine effectiveness & safety
Observational studies on COVID-19 vaccines compare event rates in vaccinated and unvaccinated person time using Poisson or Cox regression. In Cox regression, the chosen time scale needs to account for the time-varying incidence of SARS-CoV-2 infection and COVID-19 vaccination. To quantify bias in person time-based methods, with and without adjustment for calendar time, using simulations and empirical data analysis.
Health‐related quality of life the first year after a prostate cancer diagnosis a systematic review
Abstract How do patients, newly diagnosed with prostate cancer, experience their health-related quality of life? There are numerous treatment options, all affecting health-related quality of life in different ways. How each treatment method affects patients is used when guiding these patients in the choice of treatment. However, we are missing knowledge about how the newly diagnosed patient specifically experiences the first year of treatment, supporting the decision making.
