Mutant K133I of human CypD was expressed in Escherichia coli strain BL21(DE3). Bacteria were grown in Luria Broth medium at 37 °C up to an optical density (OD) of 0.6 at 600 nm and induced for 2 h with isopropyl-β-D-thiogalactopyranoside. Cells were lysed by sonication in buffer A, composed of 50 mM Tris at pH 7.5, 2 mM EDTA and 2 mM β-mercaptoethanol.
