Abstract Pears (Pyrus spp.) are self-incompatible crops with broad genetic diversity. High heterozygosity and technical limitations result in gaps within reference genomes. Our study presents the telomere-to-telomere, haplotype-resolved genomes of a representative Asian pear ‘Dangshansuli’ (Pyrus bretschneideri) and a European pear ‘Max Red Bartlett’ (Pyrus communis).

Introduction Pear (Pyrus spp.) fruits typically can be classified into green, yellow, russet brown, and red based on their peel colour (Wu et al., 2013; Xue et al., 2017), of which red cultivars account for a smaller proportion and with limited available germplasm, especially in Asian pears (Feng et al., 2010; Wu et al., 2018).

4. Materials and Methods 4.1. Plant Materials The ‘Duli’ (Pyrus betulifolia Bunge) seedlings were collected from an orchard in Hebei province, China, and were grown in a growth chamber at Nanjing Agricultural University, under a 16  h/8  h light/dark photoperiod with 75% relative humidity at 25 °C for 30 days, and used for the sugar treatments. After thirty days of treatment, the leaves of the ‘Duli’ seedlings were flash-frozen and stored at −80 °C prior to analysis.
