Integrated in vivo combinatorial functional genomics and spatial transcriptomics of tumours to decode genotype-to-phenotype relationships - Nature Biomedical Engineering
Abstract Advancing spatially resolved in vivo functional genomes will link complex genetic alterations prevalent in cancer to critical disease phenotypes within tumour ecosystems. To this end, we developed PERTURB-CAST, a method to streamline the identification of perturbations at the tissue level.
Integrated combinatorial functional genomics and spatial transcriptomics of tumors decodes genotype to phenotype relationships
Abstract Linking the complex genetic changes underlying cancer to relevant diseasephenotypes poses a challenge. Therefore, we present CHOCOLAT-G2P, a scalable approach that integrates multiplex in vivo functional genomics with spatial transcriptomics. By redeploying RNA-templated ligation probes of commercial spatial transcriptomics technology, we streamline mapping composite genetic alterations and transcriptome-wide phenotyping on the same tissue section on a single readout platform.
Phosphatidylinositol 4-kinase III alpha governs cytoskeletal organization for invasiveness of liver cancer cells

Multiplexed orthogonal genome editing and transcriptional activation by Cas12a
(a) Schematic depicting the strategy for Cas12a-VPR in which 20-nt guides produce indels and 15-nt guides allow transcriptional activation. (b) Overview of vector constructs.
GenomeCRISPR - a database for high-throughput CRISPR/Cas9 screens
The GenomeCRISPR website is available at http://genomecrispr.org and serves as main access point to the database. A main page helps users to familiarize with the database concept by providing a short description of its contents (Figure 1). There, the database can be queried by either a gene identifier or a genomic range.
