Tau isoform mapping across tissues and biofluids reveals a signature distinguishing Alzheimer’s disease from amyotrophic lateral sclerosis
Abstract Blood phosphorylated tau 217 (p-tau217) accurately detects Alzheimer’s disease (AD) pathology but is also elevated in amyotrophic lateral sclerosis (ALS), likely reflecting peripheral (big) tau released from muscle. Assays selectively measuring brain-derived (BD-) tau and BD-p-tau217 are therefore needed.
Plasma proteome profiling identified biomarkers for the differential diagnosis and molecular staging of neurodegenerative dementias
|
Abstract Blood-based biomarkers are emerging as scalable tools for the diagnosis and monitoring of neurodegenerative diseases, but markers enabling differential diagnosis across major dementias remain limited. Here we show that large-scale plasma proteomics identifies disease-associated signatures across Alzheimer’s disease, dementia with Lewy bodies and frontotemporal dementia.

SHANK3 and beta-synuclein are novel blood-based biomarkers for the Phelan-McDermid Syndrome: a pilot study
Abstract Phelan-McDermid syndrome (PMS) is a relatively frequent cause of syndromic intellectual disability (ID) and autism spectrum disorder (ASD). It is typically caused by genetic alterations in the 22q13 chromosomal region, most often involving heterozygous deletions or mutations in the SHANK3 gene. More than half of affected individuals exhibit functional impairments in speech, cognition, motor skills, and behavior.

Cortical microstructure is associated with disease severity and clinical progression in genetic frontotemporal dementia: a GENFI study - Molecular Psychiatry
Abstract The study of genetic frontotemporal dementia (FTD) allows investigating its earliest presymptomatic stages. Using cross-sectional T1-weighted and diffusion-weighted MRI, we test the hypothesis that cortical microstructural alterations, quantified as cortical mean diffusivity (cMD), are detectable earlier and are more strongly associated with clinical progression than cortical thickness (CTh).
Increased plasma DOPA decarboxylase levels in Lewy body disorders are driven by dopaminergic treatment - Nature Communications
Abstract DOPA Decarboxylase (DDC) has been proposed as a cerebrospinal fluid (CSF) biomarker with increased concentrations in Lewy body disorders (LBDs) and highest levels in patients receiving dopaminergic treatment.
Blood-based biomarkers and plasma Aβ assays in the differential diagnosis of Alzheimer’s disease and behavioral-variant frontotemporal dementia - Alzheimer's Research & Therapy
Participants were recruited at the Department of Neurology, Ulm University Hospital, within the German FTLD consortium (www.ftld.de), a quality-controlled, monitored, multicentre initiative. The study cohort comprised individuals with clinically diagnosed Alzheimer’s disease (AD; n = 18) and behavioral-variant frontotemporal dementia (bvFTD; n = 20), alongside non-neurodegenerative controls (NNC).
Neurofilaments as biomarkers in neurological disorders — towards clinical application - Nature Reviews Neurology
Abstract Neurofilament proteins have been validated as specific body fluid biomarkers of neuro-axonal injury. The advent of highly sensitive analytical platforms that enable reliable quantification of neurofilaments in blood samples and simplify longitudinal follow-up has paved the way for the development of neurofilaments as a biomarker in clinical practice.
Serum β-synuclein, neurofilament light chain and glial fibrillary acidic protein as prognostic biomarkers in moderate-to-severe acute ischemic stroke - Scientific Reports
Abstract We aimed to assess the prognostic value of serum β-synuclein (β-syn), neurofilament light chain (NfL) and glial fibrillary acidic protein (GFAP) in patients with moderate-to-severe acute ischemic stroke. We measured β-syn, GFAP and NfL in serum samples collected one day after admission in 30 adult patients with moderate-to-severe ischemic stroke due to middle cerebral artery (MCA) occlusion.
Longitudinal course of neurofilament light chain levels in amyotrophic lateral sclerosis-insights from a completed randomized controlled trial with rasagiline - Witzel - European Journal of Neurology - Wiley Online Library
INTRODUCTION Inhibiting MAO-B rasagiline reduces dopamine and serotonin catabolism, thereby increasing dopamine and serotonin availability for neurotransmission. In amyotrophic lateral sclerosis (ALS), prominent pathological involvement of dopaminergic [1, 2] and serotonergic neurons [3, 4] has been described. Moreover, rasagiline also possesses antiapoptotic and antioxidative properties [5-7], which could contribute to ALS pathogenesis [8].
Blood GFAP as an emerging biomarker in brain and spinal cord disorders - Nature Reviews Neurology
In addition to the disease-specific limitations mentioned in each section of this Review, the accurate implementation of blood GFAP measurement and the correct interpretation of the results faces other challenges (Box 1). Evidence indicates that the expression of GFAP by astrocytes increases with age in healthy individuals124, so the correlation between GFAP levels and age needs to be explored in more extensive studies to enable the definition of age-specific normal ranges.
Lipid Mediator Profiles Predict Response to Therapy with an Oral Frankincense Extract in Relapsing-Remitting Multiple Sclerosis
Abstract Lipid mediators (LMs) are a unique class of immunoregulatory signalling molecules and known to be affected by frankincense extracts. We performed LM profiling by metabololipidomics in plasma samples from 28 relapsing-remitting multiple sclerosis (RR-MS) patients who took a standardised frankincense extract (SFE) daily for eight months in a clinical phase IIa trial (NCT01450124) and in 28 age- and gender-matched healthy controls.
Author Correction: A ferroptosis-based panel of prognostic biomarkers for Amyotrophic Lateral Sclerosis
Correction to: Scientific Reports https://doi.org/10.1038/s41598-019-39739-5, published online 27 February 2019 The original version of this Article contained a typographical error in the spelling of the author Vincent Meininger, which was incorrectly given as Vincent Meninger. This has now been corrected in the PDF and HTML versions of the Article, and in the accompanying Supplementary Information file.
Neurofilaments as biomarkers in neurological disorders
Abstract Neuroaxonal damage is the pathological substrate of permanent disability in various neurological disorders. Reliable quantification and longitudinal follow-up of such damage are important for assessing disease activity, monitoring treatment responses, facilitating treatment development and determining prognosis.
Micro-droplet arrays for micro-compartmentalization using an air/water interface
Here we present a water-in-air droplet platform for micro-compartmentalization for single molecule guided synthesis and analysis consisting of a flow-system hosting dense arrays of aqueous microdroplets on a glass surface surrounded by air. The droplets are formed in a few seconds by passing a waterfront over the array of hydrophilic spots surrounded by a hydrophobic coating, thus forming a micro-droplet array (MDA).
