A lineage-specific heat-induced feedback loop controls HrcA to promote chlamydial fitness under stress
Abstract Bacterial stress responses rely on transcriptional regulation by sigma factors and repressors. In Chlamydia, which lacks a dedicated heat shock sigma factor, repressor HrcA limits chaperone gene expression under non-stressed conditions. While HrcA function may be enhanced by the chaperone GroESL in a positive feedback loop, here we identify a heat-induced negative feedback loop mediated by HagF, a protein unique to Chlamydiota and conserved across Chlamydia species.
Chlamydia plasmid-encoded protein Pgp2 is a replication initiator with a unique beta-hairpin necessary for iteron-binding and plasmid replication
Abstract The virulence plasmid of the obligate intracellular bacterium Chlamydia encodes eight proteins. Among these, Pgp3 is crucial for pathogenicity, and Pgp4 functions as a transcriptional regulator of both plasmid and chromosomal genes. The remaining proteins, Pgp1, Pgp5, Pgp6, Pgp7, and Pgp8, are predicted to play various roles in plasmid replication or maintenance based on their amino acid sequences.
Web3, We’ve Got Some Problems
2024, and everything is (seemingly) falling in favor of Web3. Following the US Securities and and Exchange Commission’s (SEC) approval of spot-ETFs for Bitcoin, Bitcoin experienced a slight plunge before surging from $46k to over $51k. Investors believe it’ll continue to see significant gains as the effects of the approval — and the upcoming “halving” event — become more apparent.
Brief Note on Smart Contract Auditing
Contrary to popular imagination, a good audit does not equate to a bug-free protocol. Getting an audit is like a test — if a protocol says it is audited, it does not mean that their security is great. Just as we don’t assume someone got a perfect score when they merely said “I got an SAT/GRE/IELTS”, we can’t assume that a protocol has achieved max-level security and effectiveness after a mere audit.
On Startups (3): The Secret Sauce
Common folklore depicts founders as gruff explorers braving the icy lashes of winter. They are lone wolves, quick to adapt to their habitats, fierce to fend off trespassers, and adamant to survive in the wilderness. Passion, the driver that propels them through storms of doubts and uncertainty, is what dictates their worth, both from their peers and from their investors. Yet is passion alone enough?
On Startups (2): What is Growth?
Evaluation (or in VC terms, due diligence) often depends on the scale of the startup and the preferences of the investor. Early stage investors, for example, will look more closely at the motivation and underlying potential due to a lack of quantifiable metrics and traction. Later stage investors have much more information to parse through, so they will naturally delve into the company’s data and reasoning process. Regardless, one factor remains true for all investors: growth, or delta.

Sectarianism: How It Arose, and How to Solve It
ShareTweetSubscribe: CDN Daily Briefing Amid the rapid rise of political sectarianism, the modern presidency has become a divider rather than a mender.
