Abstract Background Synthetic lethality (SL) occurs when an alteration in one gene renders cells vulnerable to inhibition of another, offering a strategy for targeted cancer therapy. PARP inhibitors represent the first clinically approved synthetic lethal therapy, and numerous additional candidates are now in clinical trials. Unfortunately, resistance to these therapies is common and impairs their overall effectiveness.
Abstract CREB-binding protein (CBP) is a multi-subunit scaffold protein complex in transcription regulation process, binding and interacting with ligands such as mixed-lineage leukemia (MLL) and c-Myb allosterically. Here in this study, we have revisited the concept of allostery in CBP via residue-based interaction energy calculation based on molecular dynamics (MD) simulations.
