Introduction: There is an unmet need for effective therapeutic options for patients (pts) with relapsed or refractory (R/R) cHL whose disease has progressed after a PD-1 inhibitor. Favezelimab is a LAG-3 inhibitor that had tolerable safety and effective antitumor activity when used in combination with pembrolizumab in pts with anti–PD-1–refractory R/R cHL in the phase 1/2 MK-4280-003 study (Timmerman J et al. J Clin Oncol. 2022;40(16 suppl):7545).
Aim/Background: The 9p24.1 locus is frequently amplified in rrPMBCL, leading to overexpression of PD-L1 and PD-L2 and providing a potential mechanism of immune evasion. Pembrolizumab is an anti–PD-1 monoclonal antibody that blocks the interaction between PD-1 and PD-L1 and PD-L2, enabling an antitumor immune response.
Background: The PD-L1/PD-1 pathway may be an important mechanism of tumor immune escape in hematologic malignancies. In the multicohort, phase 1b KEYNOTE-013 study of pembrolizumab (pembro) in patients with hematologic malignancies, pembro was associated with a tolerable safety profile and a promising objective response rate (ORR, 40% [4/10]) in patients with rrPMBCL.
