Dynamic regulation of lysine and arginine metabolism promotes immune evasion by limiting T cell function in cancer
Editor’s summary Abstract Supplementary Materials REFERENCES AND NOTES Information & Authors Metrics & Citations Check Access References Figures Tables Media Share Editor’s summary Metabolic adaptations alter the tumor microenvironment, but the impact on tumor progression and antitumor immunity remains unclear. Yu et al. revealed that lysine and arginine metabolism were dynamically altered with tumor progression across mouse tumor models and patient datasets.
16-h fasting optimizes cancer immunotherapy in mice and humans
Keywords diet intervention immune checkpoint therapy immunometabolism tumor metabolism tumor microenvironment Introduction The tumor microenvironment (TME) is a metabolically competitive niche where malignant cells outcompete immune cells for limited nutrients, including carbohydrates, amino acids, and lipids.1,2 During cancer progression, tumor cells exploit metabolic plasticity to upregulate diverse metabolic pathways, such as glycolysis, glutaminolysis, and lipid metabolism.3,4,5 However,...
Stereoselective C‐Aryl Glycosylation by Catalytic Cross‐Coupling of Heteroaryl Glycosyl Sulfones
Graphical Abstract Bench-stable heteroaryl glycosyl sulfones undergo desulfonylative C−C cross-coupling with aryl-substituted nucleophiles or electrophiles through two complementary catalytic systems. The reactions offer access to a wide variety of C-aryl glycoside products with high diastereoselectivity.
A Photoinduced, Nickel‐Catalyzed Reaction for the Stereoselective Assembly of C‐Linked Glycosides and Glycopeptides
Abstract A nickel catalyst mediated the union of glycosyl halides and redox-active derivatives of carboxylic acids and amines to deliver C-alkyl glycosides diastereoselectively under photoinduced conditions. This cross-coupling reaction of glycosyl nickel species with alkyl radicals generated by visible-light-promoted fragmentation of EDA complexes is amenable to the late-stage glycosylation of complex bioactive molecules and oligopeptides.
Catalytic Multicomponent Synthesis of C‐Acyl Glycosides by Consecutive Cross‐Electrophile Couplings
Abstract Reductive nickel catalysis was leveraged to orchestrate the multicomponent union of glycosyl halides, organoiodides and isobutyl chloroformate to assemble C-acyl glycosides in high anomeric selectivities. Computational studies provided insights that rationalize the origin of the high chemoselectivity. The method is applicable to the synthesis of complex C-linked disaccharides and late-stage keto-glycosylation of peptides.
