Clemastine alleviates Parkinson’s disease pathology by activating the AMPK/mTOR pathway and enhancing autophagic flux
Abstract Parkinson’s disease (PD) is a debilitating neurodegenerative disorder with no available cure, underscoring the urgent need for novel therapeutics. This study employed the comprehensive bioinformatic analyses and the experimental models to investigate the potential of clemastine (CLE) for PD treatment. Bioinformatics identified 12 distinct cell subtypes and 577 differentially expressed genes in dopaminergic neurons (DaNs), along with 167 potential PD-relevant CLE targets.
Overview of nomenclature and diagnosis of amyotrophic lateral sclerosis
Abstract The nomenclature of amyotrophic lateral sclerosis (ALS) currently is blurred, indistinct and no accurate and haven’t been properly updated since the first description, which is far from being suitable for the current implementation of clinical practise and scientific research of ALS, and urgently need an solution. Furthermore, the current diagnostic criteria need also further been improved, because the current clinical diagnosis of ALS majorly depends on the clinical manifestations yet.

Loss of TDP-43 function contributes to genomic instability in amyotrophic lateral sclerosis
1. Introduction Amyotrophic lateral sclerosis (ALS) is one of the major adult-onset neurodegenerative disorders, with a prevalence in the United States of 5 in 100,000 people (Mehta et al., 2022). The disease is characterized by the degeneration of motor neurons of the motor cortex, brainstem, and spinal cord, resulting in progressive muscle weakness, paralysis and eventual respiratory failure (Masrori and Van Damme, 2020).
