Abstract The aim of the article was to analyse the trends in oncology interventional clinical trials in the United Kingdom (UK) from 2000 to 2025, compare with major global regions, and to assess the potential impact of the New UK Clinical Trials Regulation (UK CTR) 2026 regulatory reform on the future competitiveness.
Safety and Efficacy of HexaBody-DR5/DR5 in Malignant Solid Tumors: Phase 1/2a Results and Translational Insights Into Observed Hepatotoxicity
1 Introduction HexaBody-DR5/DR5 (GEN1029) is a death receptor 5 (DR5) agonist consisting of a mixture of two non-competing humanized immunoglobulin G1 (IgG1) DR5 antibodies with an E430G hexamerization-enhancing mutation (Hx-DR5-01 and Hx-DR5-05). Hx-DR5-01 and Hx-DR5-05 recognize non-overlapping epitopes and can simultaneously bind DR5 with nanomolar affinity. The E430G Fc mutation enhances hexamer formation upon binding to DR5 on the cell surface.
Multimodal AI and tumour microenvironment integration predicts metastasis in cutaneous melanoma - Nature Communications
Abstract Accurate prognostication is essential to guide clinical management in localised cutaneous melanoma (CM), the form of skin cancer with the highest mortality. While the tumour microenvironment (TME) plays a key role in disease progression, current staging systems rely on limited tumour features and exclude key clinicopathological prognostic features.

A Phase 1/2A trial of idroxioleic acid: first-in-class sphingolipid regulator and glioma cell autophagy inducer with antitumor activity in refractory glioma | British Journal of Cancer
Study protocol MIN-001-1203 was a first-in-human, multicentric, open-label Phase I/IIA study in patients with recurrent advanced solid tumours and high-grade gliomas (HGGs; ClinicalTrials.gov identifier: NCT01792310). All enrolled patients provided written informed consent.
A phase 1b study evaluating the safety and preliminary efficacy of berzosertib in combination with gemcitabine in patients with advanced non-small cell lung cancer
Highlights • Berzosertib plus gemcitabine was well tolerated in 38 patients with advanced NSCLC. • Preliminary signs of clinical efficacy were observed. • Response rate was numerically increased in patients with high TMB and LOH scores. • Potential DDRi biomarkers should be investigated in genomically selected patients. • Berzosertib plus chemotherapy is being investigated in other treatment settings.

First-in-human study of the PARP/tankyrase inhibitor E7449 in patients with advanced solid tumours and evaluation of a novel drug-response predictor
Abstract Background This phase 1 study examined the safety, maximum-tolerated dose (MTD) and antitumour activity of E7449, a novel PARP 1/2 and tankyrase 1/2 inhibitor. Methods E7449 was orally administered once daily in 28-day cycles to patients with advanced solid tumours (50–800-mg doses).

Cancer immunotherapy and its potential cardiac complications
Recent advances in immune therapy for cancer have significantly improved the clinical outcomes of patients with advanced cancers, where prognosis has historically been very poor. With these new treatments have come new toxicities and, as the use of immunotherapy increases, we will see an increasing incidence of immune-related adverse events, with patients presenting as an emergency.
A phase Ib dose-finding, pharmacokinetic study of the focal adhesion kinase inhibitor GSK2256098 and trametinib in patients with advanced solid tumours
Abstract Background Combined focal adhesion kinase (FAK) and MEK inhibition may provide greater anticancer effect than FAK monotherapy. Methods This dose-finding phase Ib study (adaptive 3 + 3 design) determined the maximum tolerated dose (MTD) of trametinib and the FAK inhibitor GSK2256098 in combination. Eligible patients had mesothelioma or other solid tumours with probable mitogen activated protein kinase pathway activation.
Managing adverse events associated with vismodegib in the treatment of basal cell carcinoma
Basal cell carcinomas are the most common form of skin cancer. Some develop into advanced cases not suitable for standard therapy. Vismodegib is the first-in-class oral hedgehog pathway inhibitor (which is dysregulated in 90% of basal cell carcinomas), and has demonstrated efficacy for advanced disease in clinical trials. An UK expert panel met to discuss management strategies for adverse events associated with vismodegib (most commonly taste disturbances, muscle cramps and alopecia).
