Abstract X-chromosome inactivation (XCI) is essential for extraembryonic tissue growth and embryo development in mice. However, species-specific differences in its timing and regulation have left its role in human development unclear. Here, we show that the in vitro differentiation of naive human pluripotent stem cells to trophoblast stem cells and extraembryonic mesoderm cells is accompanied by XCI.
NEWS AND VIEWS 09 September 2026 A genome-rewriting approach known as adenine base editing offers a precise way to investigate gene function in early human embryos. Writing in Nature, Bower et al.1 and Jerabek et al.2 describe how they have used a precision genome-editing tool, known as base editing, in human embryos to reveal how a key gene contributes to early development, and how embryos respond to specific DNA changes.
