Inhibition of Cbl-b restores effector functions of human intratumoral NK cells

Primary human NK cells were negatively isolated from healthy donors’ buffy coats (n=6). After NK cell isolation, cells were plated in 0.2 ng/mL IL-15 supplemented NK medium and treated for 3 hours or 24 hours with Cbl-b inhibitor (3 µM). After treatment, cells were harvested and rinsed with cold PBS. Cell pellets were prepared and immediately stored at −80°C until use.
