CRISPR-Cas9 genome editing using targeted lipid nanoparticles for cancer therapy
Country singer on battle with incurable cancer at 35: ‘Starin’ into the eyes of death’ It has been one heck of a brutal year for country music and blues singer Nat Myers as he has battled cancer and also had to deal with the loss of his …
A Combinatorial Library of Lipid Nanoparticles for Cell Type‐Specific mRNA Delivery (Adv. Sci. 19/2023)
Graphical Abstract Lipid Nanoparticles In article number 2301929, Dan Peer and colleagues developed novel ionizable lipids for improved delivery of mRNA payloads. Rational lipid design together with in vivo screening identified novel lipid-based nanoparticles (LNPs) with enhanced cell trophism. This study suggests that a structural combination of lipid tail and linker can drive novel cell trophism of LNPs in vivo.
Conformation-sensitive targeting of lipid nanoparticles for RNA therapeutics
Abstract The successful in vivo implementation of gene expression modulation strategies relies on effective, non-immunogenic delivery vehicles. Lipid nanoparticles are one of the most advanced non-viral clinically approved nucleic-acid delivery systems. Yet lipid nanoparticles accumulate naturally in liver cells upon intravenous administration, and hence, there is an urgent need to enhance uptake by other cell types.
CRISPR-Cas9 genome editing using targeted lipid nanoparticles for cancer therapy
Abstract Harnessing CRISPR-Cas9 technology for cancer therapeutics has been hampered by low editing efficiency in tumors and potential toxicity of existing delivery systems. Here, we describe a safe and efficient lipid nanoparticle (LNP) for the delivery of Cas9 mRNA and sgRNAs that use a novel amino-ionizable lipid.
A modular platform for targeted RNAi therapeutics
Previous studies have identified relevant genes and signalling pathways that are hampered in human disorders as potential candidates for therapeutics. Developing nucleic acid-based tools to manipulate gene expression, such as short interfering RNAs1,2,3 (siRNAs), opens up opportunities for personalized medicine.
