Author Correction: GWAS of lifetime cannabis use reveals new risk loci, genetic overlap with psychiatric traits, and a causal effect of schizophrenia liability
Access provided by Correction to: Nature Neuroscience https://www.nature.com/articles/s41593-018-0206-1, published online 27 August 2018. In the version of this article initially published, several occurrences of the word ‘schizophrenia’ should have more accurately been worded as ‘liability to schizophrenia’ or ‘schizophrenia risk’, including in the title.
Combined cellomics and proteomics analysis reveals shared neuronal morphology and molecular pathway phenotypes for multiple schizophrenia risk genes
Abstract An enigma in studies of neuropsychiatric disorders is how to translate polygenic risk into disease biology. For schizophrenia, where > 145 significant GWAS loci have been identified and only a few genes directly implicated, addressing this issue is a particular challenge. We used a combined cellomics and proteomics approach to show that polygenic risk can be disentangled by searching for shared neuronal morphology and cellular pathway phenotypes of candidate schizophrenia risk genes.
Understanding Kafka with Factorio
Thanks to Tom de Ruijter,Steven Reitsma and Laurens Koppenol for proof reading this post. While playing Factorio the other day, I was struck by the many similarities with Apache Kafka. If you aren’t familiar with them: Factorio is an open-world RTS where you build and optimize supply chains in order to launch a satellite and restore communications with your home planet, and Kafka is a distributed streaming platform, which handles asynchronous communication in a durable way.

Genome-wide association analysis of insomnia complaints identifies risk genes and genetic overlap with psychiatric and metabolic traits
This work was funded by the Netherlands Organization for Scientific Research (NWO Brain & Cognition 433-09-228, NWO VICI 453-14-005 and 453-07-001, 645-000-003) and by the European Research Council (ERC-ADG-2014-671084 INSOMNIA).
Conditional and interaction gene-set analysis reveals novel functional pathways for blood pressure
We use GSA implemented in MAGMA (v1.07), a detailed description of which can be found in De Leeuw et al.5. Briefly, the model is based on a linear regression framework with genes as data points, with the regression equation Z=β +Bβ +Sβ +ε, with \(\varepsilon \sim {\mathrm{MVN}}\left( {0,\sigma _e^2{\hat{\mathrm \Sigma }}} \right)\). Gene p-values P are first computed from the SNP data for each gene g .
GWAS of lifetime cannabis use reveals new risk loci, genetic overlap with psychiatric traits, and a causal influence of schizophrenia
Abstract Cannabis use is a heritable trait that has been associated with adverse mental health outcomes. In the largest genome-wide association study (GWAS) for lifetime cannabis use to date (N = 184,765), we identified eight genome-wide significant independent single nucleotide polymorphisms in six regions. All measured genetic variants combined explained 11% of the variance.
Genome-wide association meta-analysis in 269,867 individuals identifies new genetic and functional links to intelligence
This work was funded by the Netherlands Organization for Scientific Research through the following grants: NWO Brain and Cognition 433-09-228 (D.P.), NWO MagW VIDI 452-12-014 (S.v.d.S.), NWO VICI 453-13-005 (D.P.), and 645-000-003 (D.P.). P.R.J. was funded by the Sophia Foundation for Scientific Research (SSWO, grant S14-27 to P.R.J.).
Item-level analyses reveal genetic heterogeneity in neuroticism
The UK Biobank Study is a major data resource, containing phenotypic measures from 503,325 participants and genetic data from 489,212 participants (July 2017 release)15. The data was released in two phases (May 2015 and July 2017), and based on this we created two separate samples on which we performed the GWA analyses.

Genome-wide association meta-analysis of 78,308 individuals identifies new loci and genes influencing human intelligence
Intelligence is associated with important economic and health-related life outcomes1. Despite intelligence having substantial heritability2 (0.54) and a confirmed polygenic nature, initial genetic studies were mostly underpowered3,4,5. Here we report a meta-analysis for intelligence of 78,308 individuals. We identify 336 associated SNPs (METAL P < 5 × 10−8) in 18 genomic loci, of which 15 are new. Around half of the SNPs are located inside a gene, implicating 22 genes, of which 11 are new findings.
Genome-wide association meta-analysis of 78,308 individuals identifies new loci and genes influencing human intelligence
Figure 3 Gene-based genome-wide analysis for intelligence and genetic overlap with other traits. (a) Negative log -transformed P values for each gene are plotted. Green dots represent significantly associated genes from GWGAS. The threshold for gene-wide statistical significant associations was set at the Bonferroni threshold of P = 2.73 × 10?6; the suggestive threshold was set at P = 2.73 × 10?5.
Happiness, health, and mortality
On the basis of a substantial dataset, Bette Liu and colleagues (Feb 27, p 874)1x1Liu, B, Floud, S, Pirie, K..., and for the Million Women Study Collaborators. Does happiness itself directly affect mortality? The prospective UK Million Women Study. Lancet. ; 387: 874–881 Summary | Full Text | Full Text PDF | PubMedSee all References1 conclude that generally feeling unhappy or stressed does not directly increase mortality in women.
