1 Introduction Nucleotides are the building blocks of nucleic acids and represent a pan-cancer metabolic dependency. Indeed, to support their uncontrolled proliferation, chemotherapy resistance, immune evasion, and metastasis, cancer cells rely heavily on hyperactive nucleotide metabolism [1, 2], exhibiting markedly elevated levels of ribonucleotides and deoxyribonucleotides by 6–11-fold and 1.25–5-fold, respectively relative to normal proliferating cells [3, 4].
