JAK2V617F reprograms Hypoxia Inducible Factor-1 to induce a non-canonical hypoxia regulon in myeloproliferative neoplasms - Leukemia
Abstract Hypoxia-inducible factors (HIFs) are master transcriptional regulators, central to cellular survival in hypoxia and frequently activated within malignancy. Whilst malignant context directs the role of HIFs within oncogenesis, these mechanisms are not well characterised. Applying the JAK2V617F myeloproliferative neoplasms (MPNs) oncogene-driver model, in which HIF-1α is stabilised in normoxia (20% O2), we sought to determine whether the modality of HIF-1 activation directs its function.
Real-world evaluation of UK high hyperdiploidy profile using a large cohort of patients provided by HARMONY data platform - Leukemia
To the Editor: The most common genetic abnormality in childhood acute lymphoblastic leukaemia (ALL) is high hyperdiploidy (HeH), accounting for 30–35% of B-cell precursor ALL cases [1]. HeH is the non-random gain of chromosomes, resulting the modal chromosome number to be between 51 and 65 (or 67) [2,3,4]. Multiple studies have associated HeH with good outcome [1, 5, 6], however, the high frequency of HeH group resulted in accounting for up to 25% of all relapses in childhood ALL [7].
Summary Background The outcome of chemotherapy in patients older than 40 years with acute lymphoblastic leukaemia is poor and myeloablative allogeneic haematopoietic stem-cell transplantation (HSCT) has a high transplant-related mortality (TRM) in this age cohort. The aim of this study was to assess the activity and safety of reduced-intensity conditioned allogeneic HSCT in this patient population.
