PTCH1 mutations and tumor-agnostic clinical outcomes with hedgehog pathway inhibitors across cancer types
Abstract Loss-of-function (LOF) alterations in PTCH1 are a hallmark of basal cell carcinoma (BCC) and drive activation of the Hedgehog (Hh) signaling pathway. Hh inhibitors are approved to treat advanced BCC, but little is known about the frequency, biology, and treatment of PTCH1 alterations in other cancers. We analyzed PTCH1 LOF alterations across diverse solid tumors and evaluated outcomes of patients with non-BCC tumors who received Hh inhibitors.

Automating Retry for Failed Terraform Launches
We work with a lot of Terraform users to orchestrate and deploy application environments. As part of the deployment process, we saw a recurring challenge among Terraform users. A number of transient errors with Terraform or the cloud service provider caused specific commands to fail. Even though simply retrying the Terraform command tends to fix the problem, it was still just as frustrating to our team as it was to the Terraform users we worked with.

Britive: Just-in-Time Access across Multiple Clouds
Traditionally when a user was granted access to an app or service, they kept that access until they left the company. Unfortunately, too often it wasn’t revoked even then. This perpetual 24/7 access left companies open to a multitude of security exploits. More recently the idea of just-in-time (JIT) access has come into vogue, addressing companies’ growing attack surface that comes with the proliferation of privileges granted for every device, tool and process.
VMware's Dev-Centered Approach to Pre-Trained Models and Generative AI
Who knows anything about pre-trained models? Who can explain how to use generative AI? Developers, platform teams, infrastructure teams, security teams — no one is that advanced. Everyone knows just a smidgeon about using pre-trained models or generative AI. Few know that a pre-trained model consists of a large data set that gets trained with machine learning. (Disclosure: I referred to ChatGPT for that definition).
Codeanywhere Founders Take on GitHub Codespaces with Daytona
Codeanywhere was one of the first web-based code editors when it was released in 2009 (as PHPanywhere). Since that time, entire developer environments have migrated to the cloud and Codeanywhere has found itself slipping behind in the ultra-competitive Cloud IDE market, where newer products like GitHub Codespaces and Replit have taken center stage. To rectify that, three Codeanywhere veterans — Ivan Burazin, Vedran Jukic and Goran Draganić — are launching a brand new company, called Daytona.
