Inhibition of cyclin-dependent kinase 8 modulates chondrocyte transition to hypertrophy and provides in vivo protection against spontaneous osteoarthritis
Abstract Chondrocyte hypertrophy is a key hallmark of osteoarthritis (OA) that drives a multitude of whole-joint disease processes. There are no approved disease-modifying pharmaceuticals in clinical use, emphasising the urgent need for innovative therapeutics. Screening of a focussed chemical library using a novel in vitro platform discovered that inhibition of CDK8/19 mediator kinases modulated chondrocyte behaviours with potential to limit OA hallmarks.
Porcupine inhibition is a promising pharmacological treatment for severe sclerosteosis pathologies - Bone Research
Abstract Sclerosteosis, an ultra-rare disorder characterised by high bone mass (HBM) and skeletal overgrowth, leads to facial paralysis, hearing loss and raised intracranial pressure, which is currently managed only through high-risk surgery. Sclerosteosis is caused by SOST mutations and loss of functional sclerostin, a protein that suppresses osteogenesis by antagonising Wnt/β-catenin signalling.

Keep your Sox on, chondrocytes!
The transcription factor Sox9 is important for cartilage formation during development, but its role in postnatal growth plates and adult articular cartilage has been uncertain. New research is revealing essential roles for Sox9 in postnatal cartilage homeostasis and in preventing post-traumatic osteoarthritis, along with new mechanisms for chondrocyte-to-osteoblast transition. Detection of early osteogenic commitment in primary cells using Raman spectroscopy
Major challenges in the development of novel implant surfaces for artificial joints include osteoblast heterogeneity and the lack of a simple and sensitive in vitro assay to measure early osteogenic responses. Raman spectroscopy is a label-free, non-invasive and non-destructive vibrational fingerprinting optical technique that is increasingly being applied to detect biochemical changes in cells.
