A conserved tyrosine motif encodes a structural vulnerability controlling stability and activity across J-domain proteins
Abstract J-domain proteins (JDPs) direct Hsp70 functions through a conserved J-domain that engages and activates Hsp70. JDPs support proteostasis and are linked to development and disease, yet how they are regulated remains unclear. Here, we identify a conserved tyrosine motif outside the canonical four-helix J-domain core, as an unexpected structural control element across the JDP family. NMR shows that phosphomimetic substitutions at these residues disrupt the J-domain fold in DNAJA1 and DNAJB1.
A multichaperone condensate enhances protein folding in the endoplasmic reticulum - Nature Cell Biology
Abstract Protein folding in the endoplasmic reticulum (ER) relies on a network of molecular chaperones that facilitates the folding and maturation of client proteins. How the ER chaperones organize in a supramolecular manner to exert their cooperativity has, however, remained unclear. Here we report the discovery of a multichaperone condensate in the ER lumen, which is formed around the chaperone PDIA6 during protein folding homeostasis.

It was a short week due to the July 4th holiday but definitely a hot one! I, for one, wasn’t prepared for it, even though it’s July and I should know better!!! The heat looks like it will break on Sunday but maybe you’re a fan of the weather. Either way, there are plenty of indoor and outdoor activities to keep everyone entertained! The Bruce Museum is a hidden gem located just up the road from us in nearby Greenwich, CT.

TGIF! The weekend is here. Happy St. Patrick’s Day! The weather is wacky again. We had some amazing days with temps in the 60’s but I hear there may be snow next week! Right now it all depends which track the storm takes. European and American models have significant differences so we shall see which is victorious. This weekend looks amazing though, so get out there and enjoy it! The Bruce Museum is a hidden gem located just up the road from us in nearby Greenwich, CT.
Weekend Guide February 5-7
Superbowl Weekend is here. Let the eating and commercial watching commence! Isn’t that the whole point, especially if your team isn’t even in it?! Punxsutawny Phil didn’t see his shadow on Groundhog’s Day indicating to the masses that Spring is right around the corner. Hopefully his expert insight is correct! I've found so many fun things to do this weekend, starting right here in Rye Brook tonight with the Blind Brook Middle School production of Joseph and the Amazing Technicolor Dreamcoat.
Weekend Guide January 29-31
The snow is behind us, hopefully for a while, and Groundhog Day is upon us next week! Will he see his shadow or not? Will we be getting 6 more weeks of winter? Who knows!? Many of us didn’t get to enjoy too many of the activities listed in the guide last weekend, so here’s to hoping you get to take advantage of all the fun stuff I found for you this weekend! The Bruce Museum is a hidden gem just up the road from us in nearby Greenwich, CT.

Much of your weekend plans will likely be altered by the impending snow storm that’s on the horizon. Are you hoping for a lot of snow or a little? Yes it’s pretty, and yes it’s been a while, and yes the kids will love but it’s also a nuisance, dangerous and messes up all our weekend plans. As you can tell, I have a love hate relationship with snow. Hopefully it won’t be too bad and you can get out and enjoy some of the fun activities I found with family and friends.
Weekend Guide January 15-18
Welcome to the three day weekend! Martin Luther King Jr. Holiday Weekend is here. As with many holidays, many of us, myself included, focus on the fact that we/the kids have a day off rather than the reason why we have the day off. On this holiday, especially in this day and time, I think it’s a good reminder to recall what Martin Luther King Jr. stood for and what his message was all about. Acceptance, Equality, and Tolerance.
Weekend Guide January 8-10
One full work/school week into the new year! Did you make resolutions? If so, are you still keeping them? Hopefully the deep freeze that had us miserable earlier this week is behind us for a little while with warmer temps giving us some thankful relief. Sunday looks like a washout so the best day for outdoor activities is most likely Saturday.
Weekend Guide January 1-3
All the holidays are behind us, for 2015, and I hope it was a good year for all of you. For those of you who had a less than wonderful 2015, try and think of 2016 as a time for new beginnings, new memories and new friends. A chance to reset. From the bottom of my heart, thank you for all your support of HamletHub throughout the year! I hope to live up to your expectations in 2016! Happy New Year.
Weekend Guide December 18-20
One week till Christmas! Many people are starting their vacation early, literally or in their minds. But with weather in the 60’s last weekend and scheduled to be in the 60s this week and maybe even hitting 70 on Christmas Eve, it’s hard for some to get in the holiday spirit. Me, I’m not complaining. I love the unseasonably high temps allowing us to be outside just a little longer before the snow and ice commence.
Weekend Guide December 11-13
Hanukkah ends this Sunday and then it’s less than two weeks till Christmas and less than three weeks till 2016. That’s insane! Despite the warm weather, we’re talking in the 60’s this weekend, it’s mid-December and Winter is officially right around the corner. Take advantage of this unseasonable warmth and enjoy some of these fun activities outdoors.
Weekend Guide December 4-6
Hanukkah arrives at sundown on Sunday. It’s a wonderful time of year for most. A stressful time of year for some too. And a hard time of year for others. The holidays bring up all sorts of emotions and can be a sad, depressing time for many. Remember to focus on what’s important and try to look ahead, not behind. I know it’s easier said than done, believe me, but with so much sadness and negativity going on around us, try to embrace and enjoy what you have today.
