Abstract Excluding false positives is critical for interpreting CRISPR screens. Here, we introduce a new Chronos module for estimating false discovery rates for identifying knockouts that cause loss of viability or have differential viability effects in different conditions. We introduce a rigorous benchmarking framework using real CRISPR data.

Abstract Proteins operate within dense interconnected networks, where interactions are necessary both for stabilising proteins and for enabling them to execute their molecular functions. Remarkably, protein-protein interaction networks operating within tumour cells continue to function despite widespread genetic perturbations.

Introduction Tumor genomes typically abound with genetic aberrations, ranging from missense mutations in individual genes to deletions of entire chromosome arms (Vogelstein et al, 2013). These genetic alterations can result in reduced functionality, or complete loss of function, of multiple proteins in any given tumor cell. Despite these alterations, tumor cells remain viable and even thrive, suggesting that they are highly robust to genetic perturbation.
