Abstract Specific inherited mutations of BRCA1 compromise homologous recombination repair and increase the risk of cancer. Here, we employ a panel of isogenic chicken DT40 cell lines for the comparative study of the main BRCA1 protein domains and of common BRCA1 founder mutations in genome stability and DNA repair.

Chain-terminating nucleoside analogs (CTNAs) have been frequently used as drugs for the treatment of virus infection and cancers [1]. CTNAs incorporated into DNA interfere with DNA replication of viruses and cancer cells. CTNAs halt replication via following two distinct mechanisms [2]. First, in the chain-termination of replication, CTNAs incorporated into the end of nascent DNA inhibit subsequent polymerization reactions.
