Adipocyte-Derived Leptolin Enhances Energy Expenditure and Prevents Obesity
1 Introduction Obesity, a chronic disease defined by excessive fat accumulation, has become an important cause of morbidity and mortality in humans [1]. Obesity results from the dysregulation of energy balance [2, 3]. Adipokines, proteins secreted by adipose tissue, play critical roles in the regulation of energy balance [4]. For instance, leptin activates hypothalamus to promote fat utilization, elevate energy expenditure (EE), reduce appetite and body weight [5-7].
The DMHGABA Neurons Play a Crucial Role in The Regulation of Cold‐Induced White Adipose Browning through DMH to LPO Projection
1 Introduction The white adipose tissue (WAT) browning can convert lipidoverloaded WAT into energy-dissipating beige adipose tissue.[1, 2] WAT browning is characterized by reduced adipocyte volume, formation of multilocular lipid droplets in white adipocytes, enhanced fat utilization, elevated energy expenditure (EE), reduced whole-body adiposity, and lowered body weight.[1, 3, 4] It is estimated that in adult humans, the browned subcutaneous fat may account for nearly 15% of resting EE or...
Age-related decline of PKA-RIIβ level in SNc dopaminergic neurons underlies PD pathogenesis
Abstract The cyclic-AMP dependent protein kinase A (Protein kinase A, PKA) regulates dopaminergic function in the substantia nigra pars compacta (SNc). However, whether PKA is involved in the pathogenesis of Parkinson's disease (PD) is unknown. Here, by collecting and analyzing the current worldwide SNc transcriptomic datasets of PD patients, we found a decline of PKA-RIIbeta subunit level in the SNc of PD patients.
Identification of leptolin as a novel anti-obesity adipokine
Abstract Adipokines are key factors in regulating energy homeostasis. We identified a novel adipokine; we named it Leptolin. In humans, leptolin levels in white adipose tissue were positively corrected with exercise, and negatively associated with body mass index. Leptolin levels were positively correlated with lipolysis-promoting gene expression. Elevated leptolin in plasma of athletes, whereas lowered leptolin in plasma of obese individuals were observed.
The discovery of phages in the Substantia Nigra and its implication for Parkinson's Disease
Abstract Background: A century ago, a mystery between virus and Parkinson's disease (PD) was described. Owing to the limitation of human brain biopsy and the challenge of electron microscopy in observing virions in human brain tissue, it has been difficult to study the viral etiology of PD. Recent discovery of virobiota reveals that viruses coexist with humans as symbionts.

ECSIT facilitates memory CD8+ T cell development by mediating fumarate synthesis during viral infection and tumorigenesis - Nature Cell Biology
Abstract Memory CD8+ T cells play a crucial role in infection and cancer and mount rapid responses to repeat antigen exposure. Although memory cell transcriptional programmes have been previously identified, the regulatory mechanisms that control the formation of CD8+ T cells have not been resolved. Here we report ECSIT as an essential mediator of memory CD8+ T cell differentiation.
Atmosphere | Free Full-Text | Projected Rainfall Triggered Landslide Susceptibility Changes in the Hengduan Mountain Region, Southwest China under 1.5-4.0 °C Warming Scenarios Based on CMIP6 Models
Abstract : Landslides are one of the most prevalent environmental disasters in the Hengduan Mountain Region. Landslides lead to severe economic damage and property loss, as well as fatalities. Furthermore, they tend to increase in the context of climate change. The purpose of this study is to comprehensively assess landslide susceptibility across the Hengduan Mountain Region in southwest China.
Succination inactivates gasdermin D and blocks pyroptosis
Fumarate targets pyroptosis A form of inflammatory cell death called pyroptosis depends on the caspase-mediated cleavage of gasdermin D (GSDMD), the fragments of which assemble into permeability pores that then kill the cell. The mechanisms regulating this important cellular process are not yet fully understood. Humphries et al. now report that the tricarboxylic acid cycle intermediate fumarate can act as an inhibitor of pyroptosis (see the Perspective by Pickering and Bryant).
Gasdermin D in macrophages restrains colitis by controlling cGAS-mediated inflammation
Abstract The functional relevance and mechanistic basis of the effects of the pyroptosis executioner Gasdermin D (GSDMD) on colitis remain unclear. In this study, we observed that GSDMD protein was activated during intestinal inflammation in a model of chemically induced colitis. GSDMD deficiency exacerbated experimental colitis independent of changes in the microbiota and without affecting the production of antimicrobial peptides.
Dopamine Uses the DRD5-ARRB2-PP2A Signaling Axis to Block the TRAF6-Mediated NF-κB Pathway and Suppress Systemic Inflammation
Highlights • Dopamine inhibits TLR2-induced NF-κB activation and inflammation via the DRD5 receptor • DRD5 can directly recruit TRAF6 and its negative regulator ARRB2 • DA-DRD5-ARRB2 signaling axis impairs TRAF6-mediated NF-κB activation and inflammation • DA-DRD5 signaling can protect mice against S. aureus-induced sepsis and meningitis Summary The functional relevance and mechanistic basis of the effects of the neurotransmitter dopamine (DA) on inflammation remain unclear.
