Abstract The evolutionary dynamics of cancer genomes at single-cell resolution under therapeutic pressure remain elusive. Hence, we perform whole-genome sequencing of 58 single-cell-derived tumoroids and 18 matched bulk tumors from six patients with metastatic colorectal cancer. High-resolution phylogenies reveal the clonal evolution of cancer with quantitative and qualitative profiling of therapy-associated mutations at the single-cell level.

Abstract Somatic cells accumulate genomic alterations with age; however, our understanding of mitochondrial DNA (mtDNA) mosaicism remains limited. Here we investigated the genomes of 2,096 clones derived from three cell types across 31 donors, identifying 6,451 mtDNA variants with heteroplasmy levels of ≳0.3%.

Abstract This retrospective study was designed to compare prognostic relevance of magnetic resonance imaging (MRI) findings before and after neoadjuvant chemoradiotherapy (CRT). From 2002 to 2010, 399 patients who underwent surgery after CRT for rectal cancer (≥T3) and had adequate pre-CRT (mr) and post-CRT (ymr) MRI findings were examined. Factors examined included tumour (T), lymph node (N), mesorectal fascia (MRF), extramural venous invasion (EMVI), and tumour regression grade (TRG).
