The absence of experimentally determined PmrB structures has significantly hindered structure-based drug design efforts and highlighted the need for accurate and reliable structural models. However, the development of accurate PmrB structure was challenged by limited sequence identity and coverage to the available homologous template in the Protein Data Bank and presents a significant challenge in structure-based drug design.
