Abstract We introduce a novel model of nonsegmented negative-strand RNA virus (NNSV) transcription. Previous models have relied on polymerase behavioral differences in the highly conserved intergenic sequences. Our model hypothesizes the transcriptional gradient in NNSVs is explained through a simple model with two parameters associated with the viral polymerase.

Abstract Biological products produced by plasmids underscore the bioeconomy, thus ensuring the sequences of physical DNA constructs match their expected designs is critical. Yet, the verification of plasmids remains a difficult and overlooked step in the production process. We developed a web application to generate certificates that embed information about a plasmid and its designer within the sequence itself.

Abstract Despite the wide use of plasmids in research and clinical production, verifying plasmid sequences is a bottleneck often underestimated in the manufacturing process. While sequencing platforms continue to improve, the chosen method and assembly pipeline still significantly influence the final plasmid assembly sequence. Furthermore, few dedicated tools exist for plasmid assembly, particularly for de novo assembly.
