WHAT IS THE CURRENT KNOWLEDGE ON THE TOPIC? In vitro studies indicated that adagrasib may be a victim of DDIs with CYP3A4 inhibitors or inducers, and a perpetrator of DDIs with CYP3A4, CYP2C9, CYP2D6, and P-gp substrates. Adagrasib has a complex metabolism profile as it is primarily metabolized by CYP3A4 following a single dose and inhibits its own CYP3A4 metabolism following multiple dosing.
