Background: Lynch syndrome (LS), an autosomal dominant genetic cancer predisposition syndrome, is caused by mutations in the DNA mismatch repair (MMR) genes MLH1, MSH2, MSH6 or PMS2. In autosomal dominant genetic cancer syndromes like LS, a germline mutation hits one MMR gene allele so that a somatic mutation in the wild-type allele of the same MMR gene will cause a complete loss of function.
