SLC25A21 regulates insulin sensitivity in subjects with obesity through ROS/AKT/KLF15-mediated alteration of GLUT4 expression
Abstract In the state of obesity, miR-548ab targets and downregulates SLC25A21, leading to reduced GLUT4 expression. However, the precise mechanism by which SLC25A21 regulates GLUT4 expression remains unclear. This study aims to investigate whether downregulation of SLC25A21 reduces GLUT4 expression by increasing reactive oxygen species (ROS) levels and inhibiting the PI3K/AKT/KLF15 signaling pathway, thereby contributing to impaired glucose metabolism.
GULP1 enhances GLUT4 translocation by counteracting ACAP1–ARF6 inhibition
Abstract The molecular underpinnings of obesity-induced insulin resistance, a key driver of type 2 diabetes mellitus (T2DM), remain incompletely understood, limiting targeted therapies. While our prior work revealed that miR‑548ab impairs glucose homeostasis by downregulating GULP1, the precise mechanism by which GULP1 regulates insulin sensitivity, particularly through the GLUT4 trafficking machinery, is unknown.
Palmitic acid promotes miRNA release from adipocyte exosomes by activating NF-κB/ER stress - Nutrition & Diabetes
Abstract The release of adipose tissue-derived miRNAs is increased under conditions of obesity, but the exact molecular mechanisms involved have not been elucidated. This study investigated whether obesity-induced increases in palmitic acid (PA) content could activate the NF-κB/endoplasmic reticulum stress (ER stress) pathway and promote the expression and release of exosomal miRNAs in adipocytes.
