To investigate the role of Notch signaling in the homeostatic skin, we treated 8- wk-old mice with a single dose of the various antagonizing antibodies (anti-Notch1, anti-Notch2, anti-Jag1, anti-Jag2, and the isotype control antibody anti-Ragweed) alone or in combination. We then examined the dorsal, resting phase (telogen) skin at 3, 7, and 14 d post-treatment. We first confirmed that our antibodies could reproduce the requirement of Notch1 in regulating sebocyte differentiation.
