Abstract Clostridioides difficile infections (CDI) cause almost 300,000 hospitalizations per year of which ~15-30% are the result of recurring infections. The prevalence and persistence of CDI in hospital settings has resulted in an extensive collection of C. difficile clinical isolates and their classification, typically by ribotype.

Abstract Analysis of the protein composition that make up C. difficile spores is essential in understanding the protein composition of its surface layers however the spore surface is composed of protein complexes hindering protein analysis. Here we compare commonly used protein extraction methods used to extract proteins of C. difficile spores.
