Integrated structural dynamics uncover a new B12 photoreceptor activation mode - Nature
Abstract Photoreceptor proteins regulate fundamental biological processes such as vision, photosynthesis and circadian rhythms1. A large photoreceptor subfamily uses vitamin B12 derivatives for light sensing2, contrasting with the well-established mode of action of these organometallic derivatives in thermally activated enzymatic reactions3. The exact molecular mechanism of B12 photoreception and how this differs from the thermal pathways remains unknown.

Engineered enzymes for enantioselective nucleophilic aromatic substitutions - Nature
Abstract Nucleophilic aromatic substitutions (SNAr) are amongst the most widely used processes in the pharmaceutical and agrochemical industries1–4, allowing convergent assembly of complex molecules through C–C and C–X (X = O, N, S) bond formation. SNAr reactions are typically carried out using forcing conditions, involving polar aprotic solvents, stoichiometric bases and elevated temperatures, which do not allow for control over reaction selectivity.
Photocobilins integrate B12 and bilin photochemistry for enzyme control - Nature Communications
Abstract Photoreceptor proteins utilise chromophores to sense light and trigger a biological response. The discovery that adenosylcobalamin (or coenzyme B12) can act as a light-sensing chromophore heralded a new field of B12-photobiology. Although microbial genome analysis indicates that photoactive B12-binding domains form part of more complex protein architectures, regulating a range of molecular–cellular functions in response to light, experimental evidence is lacking.
A Vitamin B2‐Photocatalysed Approach to Methionine Analogues
Abstract Despite their emerging role in medicinal and biological chemistry, synthetic methods to access non-natural methionine analogues remain underdeveloped. Using vitamin B2-photocatalysis, simple sulfides undergo radical α-C−H functionalization with unsaturated amino acid partners to deliver an array of new methionine derivatives. The approach also facilitates the chemoselective modification of peptide side-chains.

Synthetic Enzyme‐Catalyzed CO2 Fixation Reactions
Abstract In recent years, (de)carboxylases that catalyze reversible (de)carboxylation have been targeted for application as carboxylation catalysts. This has led to the development of proof‐of‐concept (bio)synthetic CO2 fixation routes for chemical production. However, further progress towards industrial application has been hampered by the thermodynamic constraint that accompanies fixing CO2 to organic molecules.

Enzymatic control of cycloadduct conformation ensures reversible 1,3-dipolar cycloaddition in a prFMN-dependent decarboxylase
Abstract The UbiD enzyme plays an important role in bacterial ubiquinone (coenzyme Q) biosynthesis. It belongs to a family of reversible decarboxylases that interconvert propenoic or aromatic acids with the corresponding alkenes or aromatic compounds using a prenylated flavin mononucleotide cofactor. This cofactor is suggested to support (de)carboxylation through a reversible 1,3-dipolar cycloaddition process.
Adenylation Activity of Carboxylic Acid Reductases Enables the Synthesis of Amides
Carboxylic acid reductases (CARs) catalyze the reduction of a broad range of carboxylic acids to aldehydes using the co-factors ATP and NADPH, and have become attractive biocatalysts for organic synthesis. Here we exploit our mechanistic understanding of CARs to expand their reaction scope, generating biocatalysts for amide bond formation from carboxylic acid and amine. After reaction engineering, CARs were found to have amidation activity for various acids and amines.
Mass spectrometry locates local and allosteric conformational changes that occur on cofactor binding
Native mass spectra are shown in Fig. 1, of Fdc1 Fig. 1a and Fdc1UbiXFig. 1b, sprayed from physiological-like conditions (100 mM ammonium acetate, pH 6.8). These data were used to support our previous study2. Fdc1, present only in the apo form, presents in charge states [2M+19H]19+ to [2M+23H]23+, with [2M+21H]21+ being the most intense.
