1. Introduction Drug absorption, distribution, metabolism, and excretion (ADME) are important determinants of therapeutic efficacy and safety of pharmacological treatments. This is the result of a complex interplay of cellular and molecular factors, where drug-metabolizing enzymes and transporters play a major role. The expression and activity of these proteins are far from being static or easily predictable. The same applies to the pharmacokinetics of their substrates.
