Extrapolation of lung pharmacokinetics of bedaquiline across species using physiologically‐based pharmacokinetic modelling

What is already known about this subject Bedaquiline exhibits unique pharmacokinetic characteristics, including an extremely long half-life and heterogenous caseum distribution. Bedaquiline shows extremely low unbound levels in plasma and tissues, below the minimum inhibitory concentration for Mycobacterium tuberculosis. Physiologically-based pharmacokinetic modelling enables the characterization and extrapolation of drug disposition properties in plasma and in lung tissue across species.
