Slow-SPEED: protocol for three randomised trials of remotely delivered exercise to prevent Parkinson’s disease
|
Abstract We describe the design of the first non-pharmacological Parkinson’s disease prevention trials worldwide: the randomised ‘Slow-SPEED’ trials. Three trials examine the feasibility and preliminary efficacy of a gamified, remotely administered exercise intervention vs. active control over 18–36 months in complementary prodromal subgroups: iRBD(n = 110; Netherlands), hyposmia(n = 110; United Kingdom) or LRRK2/GBA1 mutation carriers(n = 600; United States).

Combined Regulatory T-Lymphocyte and IL-2 Treatment Is Safe, Tolerable, and Biologically Active for 1 Year in Persons With Amyotrophic Lateral Sclerosis
Abstract Background and Objectives In a phase 1 amyotrophic lateral sclerosis (ALS) study, autologous infusions of expanded regulatory T-lymphocytes (Tregs) combined with subcutaneous interleukin (IL)-2 were safe and well tolerated. Treg suppressive function increased and disease progression stabilized during the study. The present study was conducted to confirm the reliability of these results.

Design of a virtual longitudinal observational study in Parkinson’s disease (AT‐HOME PD)
Abstract Objective The expanding power and accessibility of personal technology provide an opportunity to reduce burdens and costs of traditional clinical site‐centric therapeutic trials in Parkinson’s disease and generate novel insights. The value of this approach has never been more evident than during the current COVID‐19 pandemic.
Association of caffeine and related analytes with resistance to Parkinson disease among LRRK2 mutation carriers
Abstract Objective To identify markers of resistance to developing Parkinson disease (PD) among LRRK2 mutation carriers (LRRK2+), we carried out metabolomic profiling in individuals with PD and unaffected controls (UC), with and without the LRRK2 mutation.
Effect of Ezogabine on Cortical and Spinal Motor Neuron Excitability in Amyotrophic Lateral Sclerosis
Original Investigation November 23, 2020 JAMA Neurol. 2021;78(2):186-196. doi:10.1001/jamaneurol.2020.4300 Full Text Key Points Question Can ezogabine reduce cortical and spinal motor neuron excitability, which are increased in amyotrophic lateral sclerosis (ALS)? Findings In this randomized clinical trial of 65 participants with ALS, treatment with ezogabine reduced both cortical and spinal motor neuron excitability in a dose-dependent manner.

Trial of Sodium Phenylbutyrate–Taurursodiol for Amyotrophic Lateral Sclerosis | NEJM
37 References 2 Citing Articles Letters Related Articles Abstract Background Sodium phenylbutyrate and taurursodiol have been found to reduce neuronal death in experimental models. The efficacy and safety of a combination of the two compounds in persons with amyotrophic lateral sclerosis (ALS) are not known. Methods In this multicenter, randomized, double-blind trial, we enrolled participants with definite ALS who had had an onset of symptoms within the previous 18 months.

Randomized Multicenter Investigation of Folate Plus Vitamin B12 Supplementation in Schizophrenia
Importance More effective treatments are needed for negative symptoms of schizophrenia, which are typically chronic, disabling, and costly. Negative symptoms have previously been associated with reduced blood folate levels, especially among patients with low-functioning variants in genes that regulate folate metabolism, suggesting the potential utility of folate supplementation.

To the Portuguese, an apology- before I went to Lisbon, I had only the vaguest idea that Portugal even existed. It was a country I had heard of, but that was it- I had only seen it on a map. How wrong I was to have deprived myself of such a place for so long.
