Does stringent cytoreduction improve survival in advanced proliferative chronic myelomonocytic leukemia? - Leukemia
Abstract Whether mitigation of myeloproliferation improves prognosis of CMML independently of bone marrow response is unknown. Flow-defined classical monocytes (cMo) and immature granulocytes (iGRAN) have not yet been studied as biomarkers of response. We inspected the prognostic value of WBC, circulating monocytes, cMo and iGRANs in the 120 DACOTA (NCT02214407) patients randomized to decitabine (n = 63) or hydroxyurea (n = 57) evaluated after 3 cycles with BM aspiration and complete blood count.

Targeting heterochromatin eliminates chronic myelomonocytic leukemia malignant stem cells through reactivation of retroelements and immune pathways - Communications Biology
Abstract Chronic myelomonocytic leukemia (CMML) is a severe myeloid malignancy affecting the elderly, for which therapeutic options are limited. DNA hypomethylating agents (HMAs) provide transient responses, failing to eradicate the malignant clone. Hematopoietic stem cell (HSC) aging involves heterochromatin reorganization, evidenced by alterations in histone marks H3K9me2 and H3K9me3. These repressive marks together with DNA methylation are essential for suppressing transposable elements (TEs).
Eltrombopag in chronic myelomonocytic leukemia with severe thrombocytopenia. A Groupe Francophone des Myélodysplasies (GFM) study - Leukemia
Severe thrombocytopenia (<50 ×109/L) occurs in 10–30% of Chronic Myelomonocytic Leukemias (CMML) and leads to significant mortality through increased risk of bleeding and disease progression [1]. Its management is ill codified [2]. The thrombopoietin receptor agonist Eltrombopag improves platelet counts in lower-risk myelodysplastic syndromes [3]. Its activity in CMML has been investigated in retrospective case series of up to eleven patients [4,5,6], or as part of a phase I MDS trial [7].
Accelerated DNA replication fork speed due to loss of R-loops in myelodysplastic syndromes with SF3B1 mutation - Nature Communications
|
Abstract Myelodysplastic syndromes (MDS) with mutated SF3B1 gene present features including a favourable outcome distinct from MDS with mutations in other splicing factor genes SRSF2 or U2AF1. Molecular bases of these divergences are poorly understood. Here we find that SF3B1-mutated MDS show reduced R-loop formation predominating in gene bodies associated with intron retention reduction, not found in U2AF1- or SRSF2-mutated MDS.

Macrophage colony‐stimulating factor as a weapon against cytomegalovirus
Human CMV, a double-strand DNA virus of the herpes family, is a ubiquitous pathogen that is usually acquired in childhood or early adult life through contact with infected individuals' body fluids. About 70% of the population across the world carry evidence of infection. Primary CMV infection in immunocompetent people is usually asymptomatic or induces mild clinical manifestations. The virus subsequently persists lifelong in a latent form in myeloid cells of the host.
Untangling Systemic Mastocytosis: An Update on Challenges in the Diagnosis and Treatment of Myeloid Neoplasms with Systemic Mastocytosis
The remaining challenges and questions that need to be addressed for optimal treatment of SM-AHN were summarised by Radia as: what are the best approaches to risk stratification for individual patients; should we routinely use mutational profiles to guide treatment and monitor responses in all patients with SM; how and when should combination or sequential treatment be offered; and what is the place of alloHCT as a curative option?
Response to the Comments from the Groupe Francophone de Cytogénétique Hématologique (GFCH) on the 5th edition of the World Health Organization classification of haematolymphoid tumors | Leukemia
|
To the Editor: We thank Nguyen-Khac et al. [1]. who, on behalf of the Groupe Francophone de Cytogénétique Hématologique (GFCH), provide thoughtful comments on the summary of the 5th Edition of the World Health Organization (WHO) Classification of Haematolymphoid Tumors (WHO-HAEM5) [2, 3] and express that the new edition is a reflection of “the most up-to-date-knowledge on the topic”.

CDKN1A is a target for phagocytosis-mediated cellular immunotherapy in acute leukemia - Nature Communications
Abstract Targeting the reprogramming and phagocytic capacities of tumor-associated macrophages (TAMs) has emerged as a therapeutic opportunity for cancer treatment. Here, we demonstrate that tumor cell phagocytosis drives the pro-inflammatory activation of TAMs and identify a key role for the cyclin-dependent kinase inhibitor CDKN1A (p21).
The 5th edition of the World Health Organization Classification of Haematolymphoid Tumours: Myeloid and Histiocytic/Dendritic Neoplasms - Leukemia
|
Abstract The upcoming 5th edition of the World Health Organization (WHO) Classification of Haematolymphoid Tumours is part of an effort to hierarchically catalogue human cancers arising in various organ systems within a single relational database. This paper summarizes the new WHO classification scheme for myeloid and histiocytic/dendritic neoplasms and provides an overview of the principles and rationale underpinning changes from the prior edition.
