LILRB3 genetic variation is associated with kidney transplant failure in African American recipients - Nature Medicine
|
Abstract African American (AA) kidney transplant recipients exhibit a higher rate of graft loss compared with other racial and ethnic populations, highlighting the need to identify causative factors.

Multiscale mapping of transcriptomic signatures for cardiotoxic drugs - Nature Communications
Abstract Drug-induced gene expression profiles can identify potential mechanisms of toxicity. We focus on obtaining signatures for cardiotoxicity of FDA-approved tyrosine kinase inhibitors (TKIs) in human induced-pluripotent-stem-cell-derived cardiomyocytes, using bulk transcriptomic profiles. We use singular value decomposition to identify drug-selective patterns across cell lines obtained from multiple healthy human subjects.
Role of biophysics and mechanobiology in podocyte physiology - Nature Reviews Nephrology
Abstract Podocytes form the backbone of the glomerular filtration barrier and are exposed to various mechanical forces throughout the lifetime of an individual. The highly dynamic biomechanical environment of the glomerular capillaries greatly influences the cell biology of podocytes and their pathophysiology.
HCK induces macrophage activation to promote renal inflammation and fibrosis via suppression of autophagy - Nature Communications
Abstract Renal inflammation and fibrosis are the common pathways leading to progressive chronic kidney disease (CKD). We previously identified hematopoietic cell kinase (HCK) as upregulated in human chronic allograft injury promoting kidney fibrosis; however, the cellular source and molecular mechanisms are unclear. Here, using immunostaining and single cell sequencing data, we show that HCK expression is highly enriched in pro-inflammatory macrophages in diseased kidneys.

Patient cohort An overview of the discovery cohort selection process is provided in Fig. 1. We prospectively enrolled patients hospitalized with COVID-19 between March 24, and August 26, 2020, at five hospitals of a large urban, academic hospital system in New York City, NY into a cohort as previously described34.

Transcriptomic profiling of human cardiac cells predicts protein kinase inhibitor-associated cardiotoxicity
Abstract Kinase inhibitors (KIs) represent an important class of anti-cancer drugs. Although cardiotoxicity is a serious adverse event associated with several KIs, the reasons remain poorly understood, and its prediction remains challenging. We obtain transcriptional profiles of human heart-derived primary cardiomyocyte like cell lines treated with a panel of 26 FDA-approved KIs and classify their effects on subcellular pathways and processes.
Kidney tissue engineering for precision medicine
The drug development pipeline for kidney diseases is plagued with challenges ranging from an insufficient understanding of disease mechanisms to a lack of robust preclinical models. Bioengineering approaches have the potential to streamline preclinical drug discovery efforts and improve the success of clinical trials for kidney disease. References 1. Chatzimanouil, M. K. T., Wilkens, L. & Anders, H.-J. Quantity and reporting quality of kidney research. J. Am. Soc. Nephrol. 30, 13–22 (2019).
Arctigenin attenuates diabetic kidney disease through the activation of PP2A in podocytes
Abstract Arctigenin (ATG) is a major component of Fructus Arctii, a traditional herbal remedy that reduced proteinuria in diabetic patients. However, whether ATG specifically provides renoprotection in DKD is not known. Here we report that ATG administration is sufficient to attenuate proteinuria and podocyte injury in mouse models of diabetes.
