Dietary patterns in western patients with hidradenitis suppurativa: a cross-sectional study using the rapid prime diet quality screener
Abstract Hidradenitis suppurativa (HS) is a chronic inflammatory skin disease with multifactorial pathogenesis, in which dietary factors may play a role. This cross-sectional study evaluated dietary patterns in 71 HS patients in a Western population using the validated Rapid Prime Diet Quality Screener (rPDQS) and compared the results with those of a healthy historical US validation cohort representative of the Western diet.
The next generation of antibody–drug conjugates
Abstract Antibody–drug conjugates (ADCs)—composed of a monoclonal antibody linked to a payload—were designed to deliver high concentrations of cytotoxic agents to cancer cells. Since their inception, a deeper understanding of the mechanisms of action and resistance to ADCs in patients has generated a wealth of advances in the chemistry of ADC constructs, together with rational therapeutic combinations. However, the pace of innovation now exceeds clinical trial capacity.

Bridging physics and statistical learning methodologies for the accurate modeling of the radiative properties of non-uniform atmospheric paths
Satellite remote sensing is a widely spread method to characterize the state of the atmosphere and to monitor atmospheric components such as water vapor, clouds and aerosols. Atmospheric instruments measure radiances averaged over more or less narrow spectral bands. The most recent generations of satellites produce large amounts of data that need to be converted into geophysical parameters.
Helical coil design with controlled dispersion for bunching enhancement of protons generated by the target normal sheath acceleration
The quality of the proton beam produced by target normal sheath acceleration (TNSA) with high-power lasers can be significantly improved with the use of helical coils. While they showed promising results in terms of focusing, their performances in terms of the of cut-off energy and bunching stay limited due to the dispersive nature of helical coils.
The road less traveled: activating an oncogenic kinase
Under a Creative Commons license open access Keywords oncogene kinase targeted therapy PI3K Many of the most prevalent human oncogenes encode activated forms of protein or lipid kinases. This knowledge has spurred kinase inhibitor development programs that have yielded many new drugs that are now standard of care in precision oncology. Kinase inhibitors are also valuable tools for basic research.
A state-of the-art review of stratified medicine in cancer: towards a future precision medicine strategy in cancer
Highlights • Maximising outcomes with currently available targeted therapies requires a more contextualised stratified medicine • This requires consideration of the genomic, transcriptomic and immunological context of the targeted aberration • The genomic complexity and instability of human cancer requires the development of appropriate pre-clinical models • When targeting amplifications, it essential to demonstrate that any targeted amplified oncogene is an expressed driver on that amplicon...

Alpelisib Plus Fulvestrant for PIK3CA-Mutated, Hormone Receptor-Positive, Human Epidermal Growth Factor Receptor-2-Negative Advanced Breast Cancer: Final Overall Survival Results From SOLAR-1 - Annals of Oncology
Highlights • In SOLAR-1, the key secondary endpoint of OS was evaluated in patients with HR+, HER2–, PIK3CA-mutated ABC • Adding alpelisib to fulvestrant numerically improved mOS by 7.9 mo, though the result was not statistically significant • OS was numerically improved in hard-to-treat disease, including a 14-mo improvement in those with lung/liver metastases • Median time to first chemotherapy was delayed by 8.5 months with the addition of alpelisib to fulvestrant • With longer follow-up,...
Time course and management of key adverse events during the randomized phase III SOLAR-1 study of PI3K inhibitor alpelisib plus fulvestrant in patients with HR-positive advanced breast cancer
Highlights • Hyperglycemia, rash, and diarrhea were the most common grade 3/4 adverse events in patients receiving alpelisib. • These adverse events were predictable, manageable with concomitant medications, and generally reversible. • Implementation of more detailed AE management guidelines improved various markers of safety during the study.
Primary results from IMpassion131, a double-blind, placebo-controlled, randomised phase III trial of first-line paclitaxel with or without atezolizumab for unresectable locally advanced/metastatic triple-negative breast cancer
Highlights • The phase III IMpassion131 trial evaluated atezolizumab combined with paclitaxel as first-line therapy for aTNBC. • The primary endpoint was investigator-assessed PFS, tested hierarchically in the PD-L1+ and then ITT populations. • Neither PFS nor OS was improved with the combination of atezolizumab plus paclitaxel in either population. • These findings may result from imbalances in prognostic features or chance findings in a relatively small trial.
Potential blood biomarkers in chronic spontaneous urticaria
Summary Chronic spontaneous urticaria (CSU) is a mast cell‐driven disease that is defined as the recurrence of weals, angioedema or both for > 6 weeks due to known or unknown causes. As of yet, disease diagnosis is purely clinical. Objective tools are needed to monitor the activity of CSU and the efficacy of treatment. Recently, several reports have suggested that blood parameters may be considered as potential disease‐related biomarkers.
A phase III study of alpelisib and fulvestrant in men and postmenopausal women with hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) advanced breast cancer (BC) progressing on or after aromatase inhibitor (AI) the
Background: The phosphatidylinositol 3-kinase (PI3K)/mammalian target of rapamycin (mTOR) pathway is often dysregulated in HR+ BC and is associated with resistance to endocrine therapy (ET). Alpelisib (BYL719; PI3K?-specific inhibitor) and fulvestrant showed signs of antitumor activity in patients (pts) with estrogen receptor-positive (ER+), HER2– advanced BC (phase I), especially in PIK3CA-altered tumors (Janku et al. SABCS 2014, PD5-5).
Optimal strategies for the treatment of metastatic triple-negative breast cancer with currently approved agents
Triple-negative breast cancer (TNBC) is an aggressive histological subtype with limited treatment options and very poor prognosis following progression after standard chemotherapeutic regimens. Resistance to current standard therapies such as anthracyclines or taxanes limits the available options for previously treated patients with metastatic TNBC to a small number of non-cross-resistant regimens, and there is currently no preferred standard chemotherapy.
Statistical controversies in clinical research: statistical significance-too much of a good thing …
The use and interpretation of P values is a matter of debate in applied research. We argue that P values are useful as a pragmatic guide to interpret the results of a clinical trial, not as a strict binary boundary that separates real treatment effects from lack thereof. We illustrate our point using the result of BOLERO-1, a randomized, double-blind trial evaluating the efficacy and safety of adding everolimus to trastuzumab and paclitaxel as first-line therapy for HER2+ advanced breast cancer.
